Study Of Therapeutic Effect And Mechanism Of Haobie Yangyinruanjianfang On Immuno-injured Hepatic Fibrosis Induced By Bovine Serum Albumin In Rats

Objective:To study the effect of hao bie yang yin ruan jian fang on immuno-injured hepatic fibrosis induced by bovine serum albumin in rat model.To confirm therapeutic effect of hao bie yang yin ruan jian fang on experimental hepatic fibrosis and investigate the effective mechanisms,to provide experimental basis of hepatic fibrosis for clinical use of hao bie yang yin ruan jian fang.Method:The model of immuno-injured hepatic fibrosis induced by bovine serum albumin was established in Wistar rats.Bovine serum albumin containing incomplete Freund's adjuvant was first used to immunize rats(except for normal group),then through the double diffusion agar plate method to confirm whether or not BSA antibody was induced,and then by the rat tail vein to inject bovine serum albumin,2 times a week fo 15 times,thus the model was successfully established.Rats were randomly divided into 7 groups:normal group,model group,high dose of hao bie yang yin ruan jian fang,middle dose of hao bie yang yin ruan jian fang,low dose of hao bie yang yin ruan jian fang,bie jia ruan gan pian groupe,colchicine group.At the end of modeling,drugs were given to the corresponding therapeutic groups,and then were continued at the same time for two months.Distilled water was given to the rats of normal and model groups according to the same method.To count the liver weight,body weight and liver weight body weight ratio between each rat group.Liver tissues and blood serum were obtained at the end of the experiments.liver tissue pathological changes and the degree of hepatic hyperplasia were observed.Liver tissues were used for measuring the content of hydroxyproline and Liver tissue matrix metalloproteinases MMP-2,MMP-9.Blood serum were used for measuring the contents of MDA,HA,LN,?-C?PC?.Results:1 Statistics of rats in each group between the liver weight,body weight and liver weight body weight ratio:The body weight in model group was signicantly lower compared with normal group(P<0.05),and there was no significantly difference between therapy groups and normal groups.there was no significantly difference of liver weight between all groups.The liver weight body weight ratio in model group was signicantly higher compared with normal group(P<0.05),and there was no significantly difference between therapy groups and normal groups.2.Histological examination and pathological grading:The degree of hepatic fibrosis in model group was signicantly higher compared with normal groupe(P<0.05),there was no significantly difference between therapy groups and model groups,3.The contents of hydroxyproline of normal group,model group,high dose of hao bie yang yin ruan jian fang,middle dose of hao bie yang yin ruan jian fang,low dose of hao bie yang yin ruan jian fang,bie jia ruan gan pian groupe,qiu shui xian jian group were:0.4516±0.1798mg/g dry liver,1.3055±0.5048 mg/g dry liver,0.6150±0.1797 mg/g dry liver,0.6265±0.1968 mg/g dry liver,0.7186±0.2033 mg/g dry liver,0.6515±0.2265 mg/g dry liver,0.5619±0.2018 mg/g dry liver.The contents of hydroxyproline in model group was signicantly higher compared with normal group(P<0.05),and there was no significantly difference between therapy groups and normal groups.4.The contents of HA?LN IV-Cand?-C in model group was signicantly higher compared with normal group(P<0.05),and there was no significantly difference between therapy groups and normal groups.5.The contents of MDA in model group was signicantly higher compared with normal groupe(P<0.05),and there was no significantly difference between therapy groups and normal groups.6.The contents of Liver tissue matrix metalloproteinases MMP-2,MMP-9 in model group was signicantly higher compared with normal groupe(P<0.05),and there was no significantly difference between therapy groups and normal groups.Conclusion:The model of hepatic fibrosis of rat was successful,and hao bie yang yin ruan jian fang has an therapeutic effect on hepatic fibrosis induced by bovine serum albumin.The mechanism of its effects might be associated with anti-lipid peroxidation and degradation of extracellular matrix.
