Objective:To evaluate the therapeutical effect of DC targeted, nanoparticle-carried, DNA vaccine pRSC-NLDC145·gD-IL-21 in treating recurrent HSK by topical administration.Methods:Immunohistochemistry method was used for analysis of the DC targeting effect in vivo of nanoparticle-carried DNA vaccine pRSC-NLDC 145·gD-IL-21. Recurrent HSK mice model was built. C57BL/6 mice were arranged to four groups according to the DNA vaccine inoculated which were pRSC-NLDC145-gD-IL-21, pRSC-gD-IL-21, blank pRSC and blank nanoparticle-chitosan (n=6). Specific Immune reactions as targeted killing activity of spleen lymphocytes, viral neutralization antibody level in serum and the level of specific anti-HSV-1 IgA secreted in mice tears, Meanwhile, the clinical keratitis degree were evaluated.Results:The antigen of DNA vaccine pRSC-NLDC145-gD-IL-21 can be successfully targeted to corneal DC by topical administration, the specific sIgA level in teas, the viral neutralizing antibodies in serum and the cytotoxic killing activities of lymphocytes of the mice immunized with DNA vaccine pRSC-NLDC 145 gD-IL-21 were significantly increased as compared to other group mice. Most importantly, the severity of keratitis degree and the inflammatory cell infiltration in cornea were significantly decreased than other three groups.Conclusion:Topical administration of nanoparticle-carried DNA vaccine pRSC-NLDC 145-gD-IL-21 can effectively prevent or reduce the occurrence or progression of recurrent HSK in mice model.
