Sini decoction(SND)is one of the most famous and important compound in Shanghan Lun,which was composed with Radix aconite lateralis praeparata(RA),Rhizoma zingiberis(RZ)and Radix et rhizome glycyrrhize praeparata cum melle(RE).SND is excellent and compact,and modern study has indicated that SND has expressed distinct superiority and effect characteristic in prevention and clinical treatment of heart and cerebrovascular disease.RA is recognized as principle material containing alkaloids constituents.The chemical constituents of RZ are volatile oils and non-volatile constituents including diarylheptanoids and gingerol-related compounds,and the chemical constituents of RE are triterpene glycosides and flavonids.This research mainly including the following several aspects:1)Diester alkaloids are of the most important toxic components in sini decoction.At present,they were quantitative mostly in alkali conditions in High Performance Liquid Chromatographic,Or using reverse ion-pair chromatography for quantitative research,but they will cause irreversible damage to the column.Method established in this paper for the determination of Diester alkaloids,the conditions is mild and conducive to long-term use of the column.Content determination of SND only detect the glycyrrhizinate single indicator ingredients,and the ingredients of aconite and dried ginger in the herbs were not involved in 2010 version of?Chinese Pharmacopoeia?.The experiment established a method for simultaneous detectin of benzoyl new Aconitum original alkali,6-gingerol,liquiritin and the glycyrrhizic acid in SND by RP-HPLC,providing reliable detection method to control the quality of the decoction.2)Extraction process optimization of SND:?Chinese Pharmacopoeia?of the 2010 edition provide the content of SND determination only detect glycyrrhizic acid a single indicator,The experiment take benzoyl new Aconitum original alkali,licorice glycoside,glycyrrhizicacid,6-gingerol and the weight of the extract as an indicator,preferably the best boiling process.Investigating soaking time,boiling time,add water times the volume of boiling number of SND ingredients,By using traditional casserole boiling and atmospheric pressure extracting machine,and standard decoction boiling process was as follows:soak time 30 min,the boiling time 40 min,add water five times,boiling for 2 times.3)In the?Chinese Pharmacopoeia?of 2010 version,aconite in the SND prescription preparations for processed products(short Monkshood),and the prescription ratio of the light of films:dried ginger:licorice = 3:2:3,and the prescription ratio of?Shanghan Lun?(monkshood:ginger:licorice = 5:2:3)has undergone great changes,it is necessary to evaluate the efficacy of the two decoctions.In this research,mice were fed with the traditional SND and Pharmacopoeia preparations to observe the survival time of mice in hypoxic conditions and duration of ice water swimming,the results show that the traditional decoction of Sini Tang andthe Pharmacopoeia agents can significantly prolong the survival time of mice in hypoxic conditions and duration of ice water swimming.And measure the vitality of plasma SOD and MDA content in mouse plasma after the mice under hypoxic condition.Also measure the NO,ET-1 levels after ice water swimming.Results show that Mice fed with traditional SND and pharmacopoeia preparations can increase the activity of SOD in the plasma,reducing the peroxidation,make MDA content decreased,protecting the body from injury;Traditional decoction and pharmacopoeia preparations can significantly improve mouse hypoxia tolerance(P<0.01),can significantly prolong the survival time of mice under hypoxic conditions;The Traditional SND and the Pharmacopoeia preparations can significantly increased the swimming time of mice at low temperatures(p<0.01).AND can significantly prolong the survival at low temperatures.Continuous gavaging traditional SND and the Pharmacopoeia preparations,could made NO content increased and ET-1 levels to reduce in plasma of mice after ice water swimming,And the Stable blood pressure in mice may be related to the NO and ET-1.
