BackgroundOsteosarcoma is the most common malignant bone tumor among children and teenagers.It has high proliferation rate and is easy to drug resistance what lead to a high rate of mortality and morbidity in clinic. Micro RNA is a hot spot in the research field of tumor molecular biology, and plays an important role in the occurrence and development of cancer. The current study foundthat there are hundreds of Micro RNAs in serum / plasma and the specificity of these Micro RNAs with tumor disease exist certain contact. Therefore, Micro RNA has certain application value in the diagnosis, treatment and prognosis in tumor,it plays an important role in the occurrence, development and metastasis of tumor. The aim of this study is to observe the expression of Micro RNA-638 in osteosarcoma and its effect on the proliferation of osteosarcoma cell line MG63 and U20 S. Objective:To investigate the effect of Micro RNA-628 on the proliferation of human osteosarcoma cell line by observing the changes of cell proliferation in human osteosarcoma cells. Methods:(1)To detect the expression of Micro RNA-638 in the tissues and cells by quantitative real-time polymerase chain reaction after processing with 64 groups of osteosarcoma tissues and cancer adjacent tissues, and strains of osteosarcoma MG63, U2 OS, HOS, Sa0S2 cells and normal bone NH0 ST cell lines as research object.(2)To raise or lower the amount of Micro RNA-638 after Micro RNA-638 mimic or inhibitor is transfected into MG63 and U2 OS by Lipofectamine TM 2000.(3)To detect the absorbance values of 24 hs, 48 hs, 72 hs,96 hs after transfection by using ckk-8, respectively.(4)To detect the colony counts over 30 cells after 2 weeks of culture in a inverted microscope by plate clone formation assay. Conclusion:(1)In the experiment we examined the expression level of mature Micro RNA-638 in the 64 groups osteosarcoma tissues and cancer adjacent tissues by QRT-PCR, the results showed that the expression of Micro RNA-638 was down regulated in the 36 groups compared with the paired adjacent tissues. In the experiment we examined the expression level of Micro RNA-638 among osteosarcoma MG63, U2 OS, HOS, Sa0S2 cells and normal bone NH0 ST by real-time fluorescence quantitative polymerase chain reaction, the results showed that the expression of Micro RNA-638 was significantly decreased in osteosarcoma cell lines.(2)In the experiment CCK-8 and PCFA,the results showed that Micro RNA-638 could inhibit the increase of osteosarcoma MG63 and Sa0S2 cells.On the contrary, the inhibition of Micro RNA-638's expression significantly promoted the growth of osteosarcoma MG63 and Sa0S2 cells. Conclusions:The expression of miRNA-638 in human osteosarcoma tissue and osteosarcoma MG63, U2 OS, HOS and SaOS2 cell lines is low; the expression of miRNA-638 can inhibit the proliferation of human osteosarcoma cells. MiRNA-638 may become a new target for diagnosis and treatment of osteosarcoma.
