| Objective: To investigate the effect of tacrolimus, Halometasone, vitamin C possible mechanisms for the treatment of vitiligo,we use 5% hydrogen peroxide produced guinea pig model of vitiligo, then observe the melanin synthesis in skin lesions and the blood content of tyrosinaseMethods:We chose 30 normal Brown guinea pigs and they were randomly divided into 5 groups, respectively as the blank group, the tacrolimus group, the Halometasone group, tacrolimus and Halometasone group(the mixed group), and the vitaminC group. Each guinea pig was selected the back of 4x4 cm size area of skin to hair removal, with 5% hydrogen peroxide two times daily for 10 days. 10 days later, After 1 hours of hydrogen peroxide in each guinea pig, the tacrolimus group coated with 0.1% tacrolimus ointment day and night; the Halometasone group coated with 0.05% Halometasone day and night, with two weeks to stop and took one week of intermittent therapy; the mixed group coated with 0.1% tacrolimus ointment day and night, with 0.05% Halometasone at noon, Halometasone took the intermittent therapy for 2 weeks and then stopped for 1 weeks; the vitaminC group were given by gavage once a day with 90.35 mg /kg vitaminC. All the experimental groups were treated for 6 weeks.After the experiment we drew blood from the guinea pigs for testing tyrosinase content in the blood,Then we killed all guinea pigs and took the experimental area of the skin.We respectively adopted HE staining to evaluate pigmentation;Masson-Fontana to observe the melanin synthesis;Imokawa to observe the proliferation of melanocytes.Results:1.The number of melanocytes of the tacrolimus group, the Halometasone group, the mixed group were compared with the blank group of one,the difference was statistically significant(p<0.05).The number of melanocytes of the vitaminC group were compared with the blank group of one,the difference was not statistically significant(p>0.05).The number of melanocytes of the tacrolimus group and the Halometasone group were respectively compared with the mixed group of one,the difference was statistically significant(p<0.05).2.The number of melanin granules of the tacrolimus group, the Halometasone group, the mixed group were compared with the blank group of one,the difference was statistically significant(p<0.05).The number of melanin granules of the vitaminC group were compared with the blank group of one,the difference was not statistically significant(p>0.05).The number of melanin granules of the tacrolimus group and the Halometasone group were respectively compared with the mixed group of one,the difference was statistically significant(p<0.05).3.The tyrosinase content of the tacrolimus group, the Halometasone group, the mixed group were compared with the blank group of one,the difference was statistically significant(p<0.05).The tyrosinase content of the vitaminC group were compared with the blank group of one,the difference was not statistically significant(p>0.05).The tyrosinase content of the tacrolimus group and the Halometasone group were respectively compared with the mixed group of one,the difference was statistically significant(p<0.05).Conclusion:1.Halometasone, tacrolimus and Halometasone combined with tacrolimus can effectively promote melanin synthesis in the guinea pig model of vitiligo, in which the Combined use is more significant than the single use.2.Vitamin C had no inhibition on melanin synthesis of guinea pig model of vitiligo... |
