Objective:This research was conducted to analyze the differential expression of lnc RNA and m RNA within the media of vessel wall by case-control study and gain a better understanding of the molecular mechanisms in Stanford type A acute aortic dissection(AAD).Methods: Media of aortic vessel wall was collected from AAD patients(case group, group A,n=14) and normal people(control group, group C,n=10);as well as the normal media of aortic vessel wall was collected from AAD patients(self control group, group B,n=14).The lnc RNA/m RNA Human Gene Expression Microarray V4.0 was used to detect the differentially expressed lnc RNA/m RNA. Furthermore,the signal pathway and regulatory mechanism would be analysed by bio-informatics.Results: The result shows that a large number of differentially expressed lnc RNAs and m RNAs were found by comparing group A with B and A with C, HIF1A-AS1 and MEG3 were significant up-regulated. The differentially expressed m RNAs were related to 16 signaling pathways, and the significant pathways were Cytokine-cytokine receptor interaction and Jak-STAT signaling pathway.Conclusion: 1.HIF1A- AS1 is likely to interact with BRGl to inhibit vascular smooth muscle cells proliferation or promote apoptosis;MEG3 inhibit vascular smooth muscle cell proliferation and effect the extracellular matrix through TGF-? signaling pathway. 2.Cytokine-cytokine receptor interaction and Jak-STAT signaling pathway may play an important role in the molecular mechanisms of aortic dissection. 3.There are a large number of differentially expressed lnc RNA and m RNA in the aortic tissues of the patients with AAD. lnc RNA-m RNA pairs were constructed regulation network that perhaps play an important role in the disease.
