Objective: Based on the replication of rat model of asthma, we investigate its effect on the airway inflammation and liver and renal function in asthmatic rats by treating with different dosage of Asarum compatibility of Xiaoqinglong Decoction. To reveal the different dosage of Asarum compatibility of Xiaoqinglong Decoction whether there is a dose-response relationship, to reveal the safety of the Asarum and Xiaoqinglong Decoction, provides a scientific basis for the clinical safe and efficient use of Asarum.Method: 84 healthy male SD rats are fed one week after routine and divided into model group, 0 grams of Asarum compatibility of Xiaoqinglong Decoction group, 3 grams Asarum compatibility of Xiaoqinglong Decoction group, 9 grams of Asarum compatibility of Xiaoqinglong Decoction group, 15 grams of Asarum compatibility of Xiaoqinglong Decoction group, 21 grams of Asarum compatibility of Xiaoqinglong Decoction group, normal group, with 12 rats in each group. The modeling and treatment are carried out simultaneously:(1) SensitizatIon phase: Except the normal group, the other groups are injected with antigen solution 1mL(OVA 100 mg and aluminum hydroxide powder 100mg)on the first day and eighth day of the experiment, the normal group with 1m L normal saline instead.(2) Excitation and treatment phase: Sensitized 15 days later, rats are treated with different dosage of Asarum compatibility of Xiaoqinglong Decoction, the normal group and model group with normal saline instead. After filling the stomach for 1 hours, the rats are put into the self made incomplete closed glass atomization box( 60cm×50cm×50cm) to carry out atomization and then pour into the ultrasonic nebulizer 1% ovalbumin physiological saline 150 m L, with 12 rats induced at each time of atomization. Except the normal group, the other groups daily inhale 1% OVA saline excited about 20 min, 1 times a day, 14 days of continuous excitation, the normal group with normal saline instead. After the treatment, all rats are fasted for one night, collect blood samples from abdominal aorta. Measure rats addicted to eosinophil(EOS) content with automatic blood cell analyzer; Measure serum IL-5 and IL-13 content with ELISA; Measure ALT,AST,BUN,SCR content with automatic biochemistry analyzer.1 General situation of rats: in the sensitized phase of two weeks, the rats in each group are sensitive, and the skin and hair is smooth. The breathe is freely. The rats gain weight. Rats' diet and drinking water are all normal. The rats' urine and stool are also normal; On the first day of the excitation, the rats of model group and combination treatment group are scratching nose and their noses have obvious scratches. The rats have wheezing shortness of breath. The rats appeare rhythmic muscle twitching and are restless; The symptoms of model group are gradually increased with the stimulation. The rats are in bad mental state, and their skin and hair lose lustre. Rats' weight are slowly increased or even reduced; With the treatment of traditional Chinese Medicine, the rats of compound treatment group are in good mental state. Scratch nose, respiratory dyspnea and other symptoms are gradually reduced. Compared with the model group, the latent period of asthma is significantly prolonged; There is no obvious abnormal performance of normal group in the process of the whole experiment.2 Effect on the percentage of EOS in rat blood: The blood EOS percentage of normal group are lowest, and the percentage of EOS in model group are highest. There is significant difference between normal group and normal group(P<0.05);Compared with the model group, there is significant difference in the percentage of EOS in the blood of each compound treatment group(P<0.05);The blood EOS percentage of each compound treatment group is not significantly different(P>0.05),but the blood EOS percentage of 9 grams of asarum compatibility of Xiaoqinglong Decoction is lower compared with other compound treatment group.3 Effect on the content of IL-5,IL-13 in rat serum: The IL-5 and IL-13 serum content of the normal group are the lowest and The IL-5 and IL-13 serum content of the model group are the highest. There is significant difference between the normal group and the model group(P<0.05);Compared with the model group, there is significant difference in the IL-5 and IL-13 serum content of each compound treatment group(P<0.05);The IL-5 and IL-13 serum content of each compound treatment group is not significantly different(P>0.05),but the IL-5 and IL-13 serum content of 9 grams of asarum compatibility of Xiaoqinglong Decoction is lower compared with other compound treatment group. Results:4 Effect on liver and renal functions in rats: Compared with the normal group and model group, there is no significant difference in the ALT and AST serum content of each compound treatment group(P>0.05);Compared with the normal group and model group, there is no significant difference in the BUN and SCR serum content of each compound treatment group(P>0.05).Conclusion:1 Xiaoqinglong Decoction can reduce the level of blood EOS, and serum levels of IL-5 and IL-13 to inhibit the occurrence of immune inflammation of the airway in asthmatic rats.2 In accordance with the fifth edition of prescriptions dose of Xiaoqinglong Decoction, when Asarum uses from 0 g to 21 grams, other medication unchanged, they does not have obvious dose effect relationship on the inhibition of airway inflammation of OVA induced and stimulated asthmatic rats. Only 9 grams of asarum compatibility of Xiaoqinglong Decoction on the inhibition of airway inflammation of asthmatic rats is better compared with other compound treatment group.The results suggest asarum and the change of its dosage of Xiaoqinglong Decoction do not play a key role in the inhibition of airway inflammation of OVA induced and stimulated asthmatic rats.3 In accordance with the fifth edition of prescriptions dose of Xiaoqinglong Decoction, when Asarum uses from 0 g to 21 grams and other medication unchanged, Xiaoqinglong Decoction has no obvious effect on liver and kidney function(ALT, AST, BUN, SCR) of asthmatic rats.
