Objective: To study the human kallikrein binding protein in sep sis rats serum inflammatory mediators, kidney damage and renal tis sue cell apoptosis.Methods:1.Select 32 healthy clean level male SD rats, weight for 300 ± 20 g, randomly divided into 4 groups: control group(SM, n = 8), sepsis group( CP, n = 8), Ad- KS treatment group( KS, n =8), empty adenovirus treatment group( Ad, n = 8).2.Regular diet rats,After arrival to weight standard, KS group and Ad group were treated with Ad-KS and Ad through rat tail vei n injection at 48 h before cecal ligation and puncture surgery,to KS gene can be expressed in rats.3.Fasting 12 h before the cecal surgery,and from the surgery e stablish rat model of sepsis.4.From 24 h to 48 h after the surgery,blood and renal tissue wer e collected.Blood pick up from abdominal aortic for renal function examination,serum for ELISA tests.Right renal was stained by hem atoxylin eosin, and the pathological changes in renal tissue were o bserved by microscope.Left renal was collected to Western Blot met hod to detect kidney Bax/BCL- 2, the expression of KIM- 1Results:1.each group of rats renal histology changes: Group SM: Glome rular size is normal,full,the structure of renal tubular was clear,no n ecrosis,cast,did not have a large number of inflammatory cell infilt ration in renal tissue.Group CP:Multiple glomerulars were atrophy,th e structure of renal tubular was not clear,renal tubular epithelial cell necrosis,some casts had been found,a large number of inflammato ry cell infiltration in renal tissue.Group KS:Multiple glomerulars we re atrophy,the structure of renal tubular was not clear,renal tubular epithelial cell necrosis,some casts had been found,some inflammato ry cell infiltration in renal tissue,but this group was better than gr oup CP and group Ad.Group Ad was the same as group CP.2.The enzyme-linked immunosorbent assay(ELISA)method to de termine the density of blood serum concentration of TNF-? and IL-6:TNF-? and IL-6 level of group CP and group Ad significantly in creased compared with the group SM(P<0.05),TNF-? and IL-6 levelof group KS decreased compared with group CP and group Ad(P<0.05).3.Expression of apoptosis related proteins were detected by We sternblot:Expression of Bax increased in group CP and Ad compare d with group SM(P<0.05),but expression of Bcl-2 were showed n o significant deference compared with group CP,Ad and Sham.Exp ression of Bax decreased in group KS compared with CP and Ad(P<0.05),expression of Bcl-2 increased in group KS compared with gr oup CP,Ad and Sham(P<0.05).4.Expression of Kidney Injury Molecule1(KIM-1)was detected by Westernblot:In renal tissue,expression of KIM-1 in group CP a nd group Ad were significant increased compared with group SM(P<0.05).But expression of KIM-1 in group KS was significant dec reased compared with group CP and Ad.Conclusion:1.Kallistatin reduce sepsis in the rat blood serum TNF- ?, IL- 6 levels, play to the role of anti-inflammatory2.Kallistatin significantly reduce sepsis in the rat kidney tissue apoptosis, and reduce the expression of Bax/BCL-2, KIM-1, relieve kidney injury.
