Objective:The objective of this study is to identify the expression of MMP-2, TIMP-4 and there relationship with the increase of pulmonary hypertension in rats as time going on. Methods : Take 30 SPF male rats Weighs 180 grams to 200 grams and randomly divided into control group(n = 6) and the experimental group(n = 24), the experimental group give monocrotaline(MCT) 60 mg · kg-1 through intraperitoneal injection, the control group with saline volume The same weight. After 1, 2, 3, 4 weeks later,we detect the experimental group average pulmonary artery pressure and right heart hypertrophy index of rats RV/(LV + S) ratio respectively and use reverse transcription polymerase chain reaction(RT- PCR) test MMP-2、TIMP-4 mRNA expression level changes in the experimental group and control group in rats lung tissues. Expression features and location of MMP-2、TIMP-4 detected through the immunohistochemical technique.Results: Compared with control group, the rats pulmonary hypertension and right heart hypertrophy index obviously increased as time prolonged(P < 0.05), MMP-2 and TIMP-4mRNA expression quantity along with the progress of pulmonary hypertension significantly increased(P < 0.05), higher TIMP-4 compared with MMP-2, TIMP-4/MMP-2 ratio expanding.Conclusions: MMP-2 and TIMP-4’s expression in MCT induced PAH rats lung tissue obviously increased TIMP-4/MMP-2 expand ratio and balance disorders may be play an important role in the process of the lung tissue and blood vessel remodeling.
