Abstract/Summary:
In recent years, the new nano-materials because of their unique quantum size effect, volume effect and surface effect in terms of performance optics, magnetism, mechanics and other manifestations of different materials, caused a major technological change internationally. So various applications based on nanomaterials have emerged, and successfully used in the biomedical area, becoming a hot research for current scientists. In particular upconversion nanoparticles containing lanthanide can continuously absorb two or more photons of lower energy state, they emit high-energy state of a single photon, and therefore the long wavelength of near-infrared light converted into short wavelength visible light, making fluorescent nanoparticles a great potenial in biological sensing and biological imaging application. Immune chromatography (ICA) is an immunological analysis method emerged in the 1950s, immunochromatographic test strips, ICTSs effectively combined the advantages of chromatography and immunoassay, which has developed into an accurate and simple analysis techniques. This paper combined the upconversion luminescence technology with immune chromatography analysis method, used for rapid detection of macromolecular protein. The research content is as follows:In the second chapter, we constructed the upconversion luminescence technology-immuno-chromatographic analysis technology platform. Mainly from the following aspects:UCP synthesis of nanoparticles; surface modification and activation of UCP nanoparticles; the preparation of a biologically active molecule UCP-; test strip of material selection and research; the reaction principle of test strip; the preparation process of test strip. The platform is a simple, highly sensitive, versatile immunoassay platform, which can achieve rapid detection of antigen and antibody molecules, pathogens and microbes etc. Procalcitonin (PCT) is a non-hormonal activity of calcitonin (CT) propeptide substance composing by 116 amino acids with a molecular weight of 13 KD. PCT has better specificity in the diagnosis of bacterial infectious diseases, therefore it’s a highly applied-valued biological indicators in the diagnosis of infection state. In the third chapter is about converting luminescent material immunochromatographic strip in the research of detecting procalcitonin (PCT). Troponin I (cTnI) in myocardial injury occurred over six hours, we regard the cTnI as serum markers in judging the occurrence of myocardial injury. Troponin I (cTnI) with the specificity of over 96%, which has very important research value for the clinical diagnosis of acute myocardial infarction. In the fourth chapter of the thesis is about converting luminescent material immunochromatographic strip on the research of the detection of troponin I (cTnI).This subject in identifying sepsis, septicemia, bacterial and non-bacterial, systemic bacterial infections and other serious infectious diseases for early diagnosis and the rapid detection for myocardial infarction in high-sensitivity troponin, using immunochromatographic strip approach to detect, provides a reliable basis for the prevention, diagnosis and treatment of related diseases.
