Objective: To investigate the effect of Pixu 4 recipe on 5-HT,5-HT receptor,C-type natriuretic peptide,natriuretic peptide receptor and glucagon peptide 1 receptor in colonic tissue of functional diarrhea spleen deficiency syndrome rat model and explore its underlying therapeutic mechanisms.Method: Sixty three-weeks wistar rats were randomly divided into a normal control group,a model group,a positive control group and a Pixu 4 recipe cure group.Using high lactose diet and rats stood in small platform water environment to establish of functional diarrhea spleen animal model.Blood and urine samples were collected after two weeks.The content of serum lactate?the amylase detection and the excretion rate of Urine D-xylose were investigated.After modeling,the Pixu 4 recipe cure group and the positive control group were given medicine 7 days continuously,the normal control group and the model group were given normal saline.The general index and ethology of all the rats were observed during the whole experiment.After a week,the researcher collected rat colon tissue in the state of anesthesia for molecular biology and pathology specimens.The variation of 5-HT,5-HT receptor,C-type natriuretic peptide,natriuretic peptide receptor and glucagon peptide 1 receptor were detected useing immunochemistry technologies,real time PCR and Western blotting.Results: 1.The expression of 5-HT in colon tissue rats with functional diarrhea spleen was increased.The western blotting results showed that there is no significant difference on 5-HT3 receptor expression between the model group and the normal control group.5-HT4 receptor expression increased while 5-HT7 receptor expression decreased in the model group.The real-time PCR results indicated that 5-HT3 receptor expression decreased,5-HT4 receptor expression increased and 5-HT7 receptor expression decreased in the model group.After the intervention of the Pixu 4 recipe,there is no significant change on 5-HT3 receptor expression.While 5-HT4 receptor expression decreased significantly and 5-HT7 receptor expression increased significantly.2.The expression of CNP and NPR-B in colon of rats with functional diarrhea spleen decreased.After the intervention of Pixu 4 recipe,compared to the model group,there is no significant difference on CNP expression in Pixu 4 recipe group while the NPR-B expression increased.3.Compared to the control group,GLP-1 receptor expression in the colon of rats with functional diarrhea splee n decreased obviously.After the intervention of Pixu 4 recipe,Pixu 4 recipe group GLP-1 receptor expression increased obviously.Conclusion:1.5-HT and receptor may be involved in regulation of Pixu 4 recipe for functional diarrhea spleen.2.The CNP in colon and NPR-B may participate in the pathogenic progress of functional diarrhea spleen,the treatment of Pixu 4 recipe for functional diarrhea spleen through the regulation of NPR-B expression.3.Pixu 4 recipe participates in the treatment for functional diarrhea spleen by enhancing the expression of GLP-1receptor in.the colon of rats.
