Background :Atrial fibrillation(AF)is one of the most common arrhythmia clinically.The incidence is on the increase with the growth of age.It can cause a series of clinical symptoms and serious complications such as heart failure,thromboembolism events,even being disabled and death and so on severe adverse consequences.In recent years,the radiofrequency ablation(RFA)has been widely used in treating AF.However,postoperative recurrence is still the biggest challenge in treating AF at present.There has no a complete and effective method to cure the AF so far.A more effective and safer therapy approach can be obtained only by carrying out the intensive studies on pathogenesis.Multiple AF-related micro RNAs have been proved to participate in regulation in AF mechanisms currently.micro RNAs are a group of non-coded single stranded small RNA molecules which can regulate genetic expression.Related studies show that micro RNAs level changes in the circulation are associated with some heart diseases such as heart failure,myocardial infarction,coronary heart disease and myocarditis.But few studies related to regulating effect of micro RNAs in the circulation for AF have been reported at present.Investigating the changes and clinical significance of micro RNAs in the circulation in AF occurrence and progress will promote further studies in AF mechanism in order to bring in a new research direction for AF diagnosis and treatment.Objective:To investigate the changes of plasma miR-1266 and miR-150 expression levels before and after RFA;to investigate the correlation between plasma miR-1266 and miR-150 expression and different clinical AF types in order to provide a new study direction for AF diagnosis and therapeutic effect evaluation.Methods:The changes of plasma miR-1266 and miR-150 expression levels were detected in 60 patients with AF before and after RFA using real-time fluorescence quantification PCR technique as well as compared with 20 healthy PE subjects.According to AF patient's clinical types: paroxysmal AF,persistent AF and permanent AF,the changes of miR-1266 and miR-150 expression levels were compared among groups,as well as compared and analyzed with related clinical data and lab indexes.Results:1 Comparison of clinical data and lab indexes between AF group and control groupThe differences of sex,age,smoking history,left ventricular ejection fraction(LVEF),TC,TG,HDL-C have no statistical significance between AF group and control group while the differences of hypertension percentage,inner diameter of left atrium,SBP,DBP,LDL-C,Cr,BNP,hs-CRP and D-dimer have statistical significance.In the AF subgroup analysis,the differences of sex,smoking history,hypertension percentage,SBP,DBP,TC,TG,HDL-C and Cr have no statistical significance while the differences of age,inner diameter of left atrium,LVEF,LDL-C,BNP,hs-CRP,D-dimer have statistical significance.2 The difference of plasma miR-1266 and miR-150 expression in patients with AF before and after RFAThe relative expression levels of plasma miR-1266 were 1.92±0.34 and 0.16±0.06 respectively before and after RFA in the AF group.The preoperative expression increased while the postoperative expression significantly decreased.The difference was significant compared to the control group,with statistical significance(P<0.0001).The relative expression levels of plasma miR-150 were 0.35±0.06 and 1.32±0.19 respectively before and after RFA in the AF group.The preoperative expression decreased while the postoperative expression increased.The difference was significant compared to the control group,with statistical significance(P<0.0001).3 The correlation of plasma miR-1266 and miR-15060 in different types of AF patientsThe relative expression levels of plasma miR-1266 were 1.65±0.25,1.91±0.37 and 2.42±0.96 respectively in paroxysmal AF,persistent AF and permanent AF.The expression increased compared to the control group.The postoperative expression levels were 0.17±0.07,0.21±0.05 and 0.09±0.03 respectively.The expression decreased compared to the preoperative group.The difference has statistical significance(P<0.0001).The relative expression levels of plasma miR-150 were 0.17±0.07,0.21±0.05 and 0.09±0.03 respectively in paroxysmal AF,persistent AF and permanent AF.The expression decreased compared to the control group.The postoperative expression levels were 1.53±0.14,1.67±0.21 and 0.51±0.13 respectively.The expression increased compared to the preoperative group.The difference has statistical significance(P<0.0001).Conclusion1 The expressions of plasma miR-1266 and miR-150 in patients with AF are different before and after RFA.The expressions of plasma miR-1266 increased in AF group compared to the control group before operation while it significantly decreased compared to the control group after operation.The expressions of plasma miR-150 decreased in AF group compared to the control group before operation while it significantly increased compared to the control group after operation.It indicates that plasma miR-1266 and miR-150 have participated in regulation and very close to AF mechanism.It provides an extensive research direction for AF treatment and prognosis.2 The changes of plasma miR-1266 and miR-150 of expression in patients with paroxysmal AF,persistent AF and permanent AF are different before and after RFA.It indicates that plasma miR-1266 and miR-150 are closely associated with AF progression.
