Objective:To investigate the expression of the key factors which regulate bone resorption process,osteoprotegerin(OPG),ligand of receptor activator of NF-?(RANKL)and receptor activator of NF-?(RANK)in the epithelium tissue of dentigerous cyst,keratocystic odontogenic tumor and amelobblastoma,compared with the normal membrane,and to discuss the difference and its significance.Methods:Immunohistochemistry SP staining was used to evaluate the expression grade of RANKL,RANK and OPG in 69 cases of dentigerous cyst,keratocystic odontogenic tumor,and amelobblastoma specimen(26,23,20 respectively).10 cases of normal mucosa were used as control group.All those cases of the disease were confirmed by pathological examination.Results:1 The strong positive ratio of RANKL in 69 cases of disease was 68.1%.Negative ratio was 14.5%.There were significant differences between the diseases and normal mucosa(P<0.05)in expression of RANKL,and for further step analysis showed that there was no significant(P>0.05)difference among three type diseases.2.There was no case showed strong positive expression of OPG in 69 cases of disease.The positive ratio of normal mucosa was greater than other groups.After statistics analysis,we have drawn a conclusion that the sequence of the expression grade of OPG in three diseases should be listed as follows: normal mucosa>keratocystic odontogenic tumor>amelobblastoma>dentigerous cyst3.There were no meaningful differences in expression grade of RANK among the four groups.(P>0.05)4.Keratosis cystic tumors expressing OPG/RANKL/RANK :strong positive expression rate of OPG/RANKL/RANK were 0.0%,69.6%,39.1%,highest expression levels of RANKL;weakly positive expression rate respectively were85.6%,,26.1%,34.8%.Analysis shows that a higher level of expression of RANKL and OPG in keratocystic odontogenic tumor group.There was a significant positive correlation(spearman rank correlation test)between RANKL and RANK,there is no correlation between OPG and RANK or RANKL5.The expression of OPG / RANKL / RANK in dentigerous cyst is: RANK,RANKL,OPG strong expression was 50.0%,73.1%,0%.While weakly positive expression of RANK / RANKL / OPG were 30.8%,11.5%,61.5%.In dentigerous cyst also showed high expression levels of RANKL,but OPG expression levels are relatively low.There was a significant positive correlation(spearman rank correlation test)between RANKL and RANK,there is no correlation between OPG and RANK or RANKL.6 The expression of OPG / RANKL / RANK in: RANK,RANKL,OPG strong positive rate of 35%,60%,0%,weakly positive were 50%,15%,40%,that enamel ameloblastoma has a higher levels of RANKL expression.There was a significant correlation between RANKL?RANK and OPG.Conclusion:In the normal mucosa,dentigerous cyst,keratocystic odontogenic tumor,and ameloblastoma tissue,the expression of OPG and RANK,RANKL can be detected.From the experimental data,it is shown that the OPG expression in the three diseases are different,and RANKL and RANK expression between the three groups of disease does not exist significant,we hypothesized that OPG may play a major role in dentigerous cyst and keratocystic odontogenic tumor,ameloblastoma for bone resorption differences.
