Objective: To explore the effect of Notch signaling pathway silencing on EMT and molecular mechanism with Bladder Cancer in Vivo.Methods: 5637 human Bladder tumor xenongrafts were used as preclinical tumor models?The Bladder tumor nude mice(15)were divided randomly into three groupsand : dosing group(gamma-secretase inhibitor treatment group),the transfection group(silence Notch-1 receptor group)and control group.The skin measuring size of the tumor were measured every 7 days,After 5 weeks,the mice were sacrificed and the weigh and size of the tumor were measured.Nature of the tumors had pathological detection,immunohistochemical detection of tumors in EMT marker protein expression,by RT-PCR and for westen blot technique to detect each group within tumors had organization E cadherin,N-cadherin,vimentin m RNA and protein expression changes.Results: Dosing group tumors growth rate was significantly lower than control group(P < 0.05),fluid transfection group growth rate is lower than the control group(P < 0.05),dosing tumors growth rate is lower than the transfection group(P < 0.05),after five weeks,dosing tumors had the size,quality is significantly lower than control group(P < 0.05),transfection group tumors had the size,the quality is lower than the control group(P < 0.05),dosing tumors had the size,the quality is lower than the transfection group(P < 0.05),the pathological confirmed hematoxylin-eosin staining for malignant tumor,microscopically,the tumor cell sizes,hyperchromatic nuclei,cytoplasm is rich,alien,cell density.By immunohistochemical technique to detect tumors had the EMT marker protein expression of the situation,the result shows: the transplanted tumor in tumor tissues,transfection group E cadherin,N-cadherin protein expression level,vimentin protein expression down;Dosing group E-cadherin protein expression level,and N-cadherin,vimentin protein expression cut(figure 15).By rt-pcr and western blot technique to detect the tumors had the organization E cadherin,N-cadherin,vimentin m RNA and protein expression changes,the result shows: the transfection group E cadherin,N-cadherin m RNA and protein expression significantly higher(P<0.05),vimentin m RNA and protein significantly lowered(P<0.05);Dosing group E-cadherin m RNA and protein expression significantly higher(P < 0.05),while N-cadherin,vimentin m RNA and protein significantly lowered(P<0.05).Conclusion: In nude mice in vivo,through silence Notch signal pathway,can effectively inhibit bladder EMT process;At the same time reduce bladder cancer cell apoptosis resistance and aggressive.
