Background:Gastric cancer is a malignant tumor of gastric mucosal epithelium and glandular epithelium, which is one of the common tumors in the digestive system, and is a kind of malignant tumor with high mortality. At present, chemotherapy is an important treatment for gastric cancer, cisplatin (DDP) is one of the most commonly used drugs for treatment of gastric cancer. However, it is easy to produce drug resistance to chemotherapeutic drugs. Therefore, it is very important to find a kind of chemotherapeutic agent to improve the efficacy of chemotherapy in the treatment of gastric cancerMicroRNA (miRNA) is a kind of small molecule RNA with a length of about 20nt, it plays an important role in the development of a variety of tumors including gastric cancer by exerting the role of oncogenic factor or tumor suppressor factor. It has been confirmed that miRNA is involved in the regulation of tumor resistance to chemotherapy, and provides a new idea for the development of new chemotherapeutic agents.The research group according to the gastric carcinoma and adjacent normal tissue samples analysis of microarray results, there are many different expression of miRNAs in gastric carcinoma, our study was significantly downregulated miR-125b.Objective:To investigate the mechanism of microRNA-125b (miR-125b) in gastric cancer and its effect on DDP.Methods:Quantitative reverse-transcriptase polymerase chain reaction was used to detect the expression of miR-125b in gastric cancer and adjacent normal tissues. The migration and invasion ability was detected by cell migration assay and cell invasion assay in miR-125b overexpression and Negative Control (NC) of gastric cancer cells. The proliferation of gastric cancer cells in two groups of gastric cancer cells was detected by cell proliferation assay.Results:The expression of miR-125b in gastric cancer tissues was significantly lower than adjacent normal tissues. The migration and proliferation of miR-125b overexpression gastric cancer cells were significantly decreased, and the proliferation ability of the treated group by DDP was obviously decreased. In the correlation analysis of miR-125b expression and clinical indexes of patients found that miR-125b expression was related to lymph node metastasis, HER2 expression and overall survival of patients, and the difference is statistically significant (P< 0.05).Conclusion:miR-125b plays an important role in gastric cancer. It inhibits the proliferation and metastasis of gastric cancer cells, and increases the chemosensitivity of gastric cancer cells to DDP.
