BackgroundThyroid gland is made up of 3 million follicles,and it is bigger than other endocrine glands of human. The diameter of follicles is from 15 to 500 microns. Thyroid gland is the only endocrine gland which stockpiles its product outside the cells. The thyroid hormone not only promotes the growth of our body, but also adjusts the energy and substance metabolism of our body. In addition, it can influence the organ function.Thyroid carcinoma is the fastest-growing solid malignancy, accounting for about 1.5% of all malignant tumors. Since the 21 st century, the patients of thyroid cancer are sudden rising. In China, it has ranked fourth in female malignancy and taken the first place in Korea. As the most common type, papillary thyroid carcinoma has the best prognosis, but the molecular is not clear now.Annexins is a kind of phospholipid binding protein. It can not exert biological functions without calcium ion. Annexins involve the construction of ion channel, the formation of bioflim and signal transduction. They also have relationships with endocytosis, exocytosis, cell proliferation and apoptosis and cellular differentiation. Both ANXA2 and ANXA4 are two important members of the family. The abnormal expressions of ANXA2 and ANXA4 are related to the occurrence and progression of many malignancies. To research the role of ANXA2 and ANXA4 in thyroid tumor, immunohistochemistry, RT-PCR and Western-Blot were used to detect the protein and m RNA expression of ANXA2 and ANXA4 in PTC, corresponding to non-tumor normal tissues and benign thyroid tumor and their correlation with clinicopathologic characteristics were analyzed. We look forward to lying the foundation of the diagnosis and treatment of PTC. ObjectiveWe detect the protein and m RNA expression of ANXA2 and ANXA4 in PTC, corresponding non-tumor normal tissues and benign thyroid tumor and their correlation to clinicopathologic characteristics were analyzed. Furthermore, we explore the functions of ANXA2 and ANXA4 in thyroid tumor. MethodsThe tissues were gathered from the First Affiliated Hospital of Zhengzhou University, from June 2014 to April 2015. All the tissues were verified the pathological patterns by clinical pathologist.The tissues contain 78 cases of PTC, 35 cases of benign thyroid tumor and 20 cases of normal thyroid tissue. We use the immunohistochemistry, RT-PCR and Western-Blot methods were used to study the differential expression of ANXA2 and ANXA4 in different thyroid tissues. The statistical approach is utilized to analysis the relationships between the two indexes and clinicopathological parameter of PTC. We utilize SPSS 18.0 to deal with experimental datum. ResultThe results of ANXA2: There are 56 positive ANXA2 protein expression in 78 samples of PTC, 4 positive expression in 35 samples of benign thyroid tumor, only 1 positive expression in 20 samples of normal tissue. The chi-square is 51.29 and P is less than 0.01. The results are statistically significant.The relative expressions of ANXA2 m RNA in PTC, benign thyroid tumor and normal tissues are 0.578±0.197,0.151±0.046,0.112±0.034. One-way variance analysis illustrates significance with P<0.01. The relative expressions of ANXA2 protein in PTC, corresponding non-tumor normal tissues and benign thyroid tumor are 0.762±0.133,0.233±0.109,0.217±0.084. One-way variance analysis illustrates significance with P<0.01.The ANXA2 expression is associated with the patient's pathological TNM stages.The results of ANXA4: There are 60 positive ANXA4 protein expression in 78 cases of PTC, 3 positive expression in 35 case of benign thyroid tumor, 2 positive expression in 20 samples of normal tissue. The chi-square is 59.41 and P is less than 0.01. The results are statistical significant.The relative expressions of ANXA4 m RNA in PTC, benign thyroid tumor and normal tissue are 0.641±0.154,0.266±0.082,0.188±0.034. One-way variance analysis illustrates significance with P<0.01. The relative expressions of ANXA4 protein in PTC, benign thyroid tumor and normal tissues is 0.832±0.307,0.337±0.128, 0.312±0.089. One-way variance analysis illustrates significance with P < 0.01.The ANXA4 expression is associated with the patient's diameter of tumor and pathological TNM stages. Conclusion1. The higher expression of ANXA2 and ANXA4 may play an important role in the carcinogenesis and progression of PTC. They maybe cancer-promoting genes of PTC, and potential markers in the diagnosis and gene treatment of PTC.2. ANXA2 and ANXA4 are potential markers in diagnose assessment and illness monitoring of PTC.
