Objective:The aim of this study is to assess the association between polymorphisms of HLA-A, HLA-B and allergy, by measuring HLA-A and HLA-B alleles in patients with succinylated gelatin allergy.Methods:Clinical criteria for diagnosing of succinylated gelatin allergy: after exposure to succinylated gelatin, a few minutes or hours appear at least two or more of the following symptoms(1) skin reactions, such as skin flushing, maculopapular rash and hives;(2) respiratory system symptoms, such as bronchospasm and hypoxemia;(3)decreased blood pressure and related symptoms, such as shock and syncope;(4)continuous gastrointestinal symptoms, such as gastrointestinal cramps and vomiting.And the allergic reaction is divided into 4 levels, the second level or above(not including the grade II) is defined as a severe allergic reaction.We performed an association study of 12 subjects with succinylated gelatin allergy, 572 general subjects from the human MHC database, all of whom are Han Chinese. Serum tryptase levels were examined using enzyme-linked immunosorbent assay(ELISA) and the frequencies of HLA-A and HLA-B alleles using polymerasechain reaction with sequencespecific oligonucleotides(PCR-SSO) combined with flow cytometry technique.Results:The frequency of the succinylated gelatin allergy patients carrying the HLA-A*01:01?HLA-B*44:02 and HLA-B*46:01 alleles are 17%?25% and 17%.They are significantly higher compared with the general Chinese Han population(2%?0% and 2%). The frequency of the succinylated gelatin allergy patients(class?above) carrying the HLA-A*31:01 ? HLA-B*44:02 ? HLA-B*46:01 alleles are33.3%?50% and 33.3%. Which are significantly higher compared with the general Chinese Han population(4%?0% and 1%).Conclusion:These findings suggest that in the Chinese Han population, HLA*31:01 ?HLA-B*44:02 and HLA-B*46:01 maybe associated with severe succinylated gelatin allergy.
