Background and Objective:Multiple sclerosis(sclerosis multiple, MS) is a chronic inflammatory disease of the central nervous system with unknown etiology, which is characterized by the spatial and temporal characteristics. For the etiology of MS, there is currently no definitive studies, epidemiological study had shown MS is a multiple factors disease, which immunomodulatory mechanism disorder plays an important role in the pathogenesis. Recent studies have found that vascular endothelial growth factor(Vascular Endothelial Growth Factor,VEGF)which involved in the destruction of the blood brain barrier, angiogenesis has an important role in the course of the MS disease,and may be the main cause of chronic inflammatory MS disease.Micro RNA(micro RNA, miRNA) is a kind of short chain non coding RNA which can express in human body. It plays an important role in cell proliferation, differentiation, apoptosis and angiogenesis. Recent studies have shown that a variety of stable expression of miRNA can be detected in the peripheral circulation of MS patients, and there are significant differences in miRNA expression between normal controls and patients. Therefore, it is concluded that the change of the expression of miRNA in the peripheral circulation has important value for the diagnosis and differentiation of MS.Our research focus on the expression of miR-199a-5p ?miR-126?miR-200 c and VEGF between the patients of multiple sclerosis in relapse and remission with healthy control.Analysis the expression differences of miR-199a-5p?miR-126 and miR-200 c in MS,and then explore their relationships in the course of multiple sclerosis. Methods:1?With 45 cases of multiple sclerosis patients in relapse group, 37 cases of multiple sclerosis patients in remission group and 40 healthy volunteers in control group,using the method of Enzyme-linked immunosorbent assay(ELISA),the changes expression level of VEGF in the three group were detected.2?With 45 cases of multiple sclerosis patients in relapse group, 37 cases of multiple sclerosis patients in remission group and 40 healthy volunteers in control group,using the method of q PCR,the changes expression level of miR-199a-5p?miR-126 and miR-200 c in the three group were detected. Results:1?The serum expression level of VEGF in relapse group was 50.75±11.31,and it was 27.36±9.83 in remission group while it was 15.64±3.26 in control group.The serum expression level of VEGF in relapse and remission group were both higher than that in control group, the differences was significant(P<0.05).The serum expression level of VEGF in relapse group was higher than remission group,the difference was significant(P<0.05).2?The serum expression level of miR-199a-5p in relapse group was 0.54±0.13,and it was 0.73±0.22 in remission group while it was 0.74±0.39 in control group.The serum expression level of miR-199a-5p in relapse group was lower compared with control group,the difference was significant(P<0.01).The serum expression level of miR-199a-5p in remission group was lower compared with control group,but the difference was not statistically significant(P=0.235).The serum expression level of miR-199a-5p in relapse group was lower compared with remission group,the difference was significant(P=0.001).3?The serum expression level of miR-126 in relapse group was 0.68±0.24,and it was 1.29±0.61 in remission group while it was 1.04±0.58 in control group.The serum expression level of miR-126 in relapse group was lower compared with control group,the difference was significant(P<0.001);The serum expression level of miR-126 in remission group was higher compared with control group,the difference was significant(P=0.003);The serum expression level of miR-126 in relapse group was lower compared with remission group,the difference was significant(P<0.001).4?The serum expression level of miR-200 c in relapse group was 2.26±0.75,and it was 0.99±0.57 in remission group while it was 0.76±0.42 in control group.The serum expression level of miR-200 c in relapse group was higher compared with control group,the difference was significant(P=0.005);The serum expression level of miR-200 c in remission group was higher compared with control group,the difference was significant(P<0.001);The serum expression level of miR-200 c in relapse group was higher compared with remission group,the difference was not statistically significant(P=0.062).5?The negative correlation were both found in the serum expression of relapse group and remission group between miR-199a-5p and VEGF(rrelapse=-0.929,prelapse=0.002;rremission=-0.955,premission=0.000).6?The negative correlation was found between miR-126 and VEGF in the serum expression of relapse group(r=-0.601,p=0.001);the positive correlation of that was found in the remission group(r=0.893,p=0.000).7?The positive correlation were both found in the serum expression of relapse group and remission group between miR-200 c and VEGF(rrelapse=0.81,prelapse=0.005;rremission=0.843,premission=0.000). Conclusion:1?The expression of miR-199a-5p is down-regulated in the serum of MS patients.2?The expression of miR-200 c is up-regulated in the serum of MS patients.3?The expression of miR-126 are different in the serum of MS patients with different course of disease,it is down-regulated in the relapse stage while it is up-regulated in the remission stage.4?VEGF is related to the pathogenesis of MS, the higher the level of VEGF,the more possibility of MS tend to be relapse.5?The level of VEGF is down-regulated by miR-199a-5p, up-regulated by miR-200 c, and up or down regulated.by miR-126. So it may induce the incidence of MS.Among them,miR-199a-5p and miR-126 may be used as a diagnostic indicator of MS for the differentiation of relapse stage.miR-200 c may be used as a possible diagnostic indicator of MS.
