ObjectiveMyasthenia Gravis(Myasthenia Gravis, MG) is a kind of autoimmune diseases, repeatedly striated muscle weakness as clinical manifestations. About 80% to 90% MG patient have acetylcholine receptor antibody(AchRab) in the serum, AchRab combine with acetylcholine receptor in the neuromuscular synaptic membranes,mainly through competitive occupation and complement mechanism lead to the nerve impulse can not download cause the symptom of muscle weakness.About 70% MG patients with thymic pathological abnormalities, of which 10% to 15% of patients with thymoma, 50% to 60% of thymic hyperplasia. Professor Zhang Qingyong findings of thymoma and thymic hyperplasia tissue acetylcholine transferase expression increased,Thymic hyperplasia tissue acetylcholine receptor expression is higher than patients with thymoma thymus tissue,Thymic atrophy organization acetylcholine receptor expression under normal thymus tissue [1]. The study found immune cells and epithelial cells express non-neuronal cholinergic system, the composition mainly arecholine acetyltransferase(CHAT), acetylcholinesterase(AchE), nAchR(?1-?10, ?1-?4, ?), mAchR(m1-m5), acetylcholine, etc. [2]. Foreign researchers found that stimulation nAchR and mAchR can generate a lot of changes with biology and function [3]. We hope that by studying the MG thymus tissue in patients with non alter neuronal cholinergic system, and to explore its effect on acetylcholine receptor antibody production. ObjectiveClinical diagnosis and surgical removal of the thymus of MG patients 30 cases, including 18 males and 12 females, aged 18 to 42 years; the control group non-autoimmune thoracotomy patients with congenital heart disease in 20 patients, aged 2 to 12 years old. Obtain the consent of patients and their families when collecting specimens, and all patients signed informed consent. MethodsCollecting 15 thymus tissues of myasthenia gravis patients(pathological characteristics was thymic hyperplasia) from the chest surgery of the second affiliated hospital of Zhengzhou University. Setting them as MG group; 15 thymus tissues of no autoimmune disease but congenital heart disease patients,setting them as normal control group.1. m1, m2, m3, m4, m5, ?1, ?2, ?3, ?4, ?5, ?6, ?7, ?9, ?10, ?1, ?2, ?3, ?4, CHAT, mRNA detection AchE of: extracting MG patients and normal thymus tissue total RNA by quantitative PCR m1, m2, m3, m4, m5, ?1, ?2, ?3, ?4, ?5, ?6, ?7, ?9, ?10, ?1, ?2, ?3, ?4, CHAT, mRNA expression levels of AchE, using 2- ? ? Ct comparative Ct method between MG patients and normal control group relative quantification;2. ?1, ?2, ?9, m3 and ?2 molecular distribution and expression in thymus cells: making thymus tissue paraffin sections by immunohistochemistry understand ?1, ?2, ?9, m3 and ?2 distribution and expression in thymus tissue;3. Statistical analysis: SPSS 21..0 statistical software for statistical analysis. Data measurement data with mean ± standard deviation( SX±), said the two groups were compared by t-test, when p <0.05 considered statistically significant when. Results1. m1, m2, m3, m4, m5, ?1, ?2, ?3, ?4, ?5, ?6, ?7, ?9, ?10, ?1, ?2, ?3, ?4, CHAT, AchE mRNA expression in thymus tissue: Real PCR results showed that compared with normal thymus ?1, ?9, express m3 thymus in myasthenia gravis patients was significantly higher(p <0.05), ?2, ?2 expression in patients with myasthenia gravis, the thymus was significantly lower(p <0.05), ?3, ?5, ?6, ?7, ?10, ?1, ?3, ?4, ?, CHAT, no difference of m1, m2, m4, m5 expression.2. ?1, ?2, ?9, m3 and ?2 expression and distribution of molecules in thymocytes, ?1, ?2, ?9, m3, ?2 expression was brown granules were expressed mainly in the cytoplasm and cell membrane, and occasionally The expression in the nucleus. ?1 expression in MG thymus tissue positive expression was positive in the control group, ?1 expression control group(p <0.05) in MG thymus tissue. ?2 expression in MG thymus tissue weakly positive expression was positive in the control group, ?2 expression in MG thymus tissue than the control group(p <0.05). ?9 expression in MG thymus tissue weakly expressed weakly positive in the control group, and the expression was no difference(p> 0.05) in the control group MG thymus tissue. m3 expressed in MG thymus tissue positive expression was positive in the control group, and the expression was no difference(p> 0.05) in the control group MG thymus tissue. ?2 expression in MG thymus tissue positive expression was positive in the control group, in MG thymus tissue was lower than that in the control group(p <0.05). Conclusions?1 expression levels in patients with MG thymus tissue was significantly increased, mRNA expression levels of ?2, ?2 is significantly reduced. ?1??2 and ?2 expressed in the thymus tissue of patients with myasthenia gravis were positive, mainly distributed in the thymus cell membrane and cytoplasm.
