Effects Of Tectoridin, A Blackberry Lily Extract Of Chinese Traditional Medicine On The Suppression Of The Proliferation And Invasion In Human Colon Cancer Cells

Objective To investigate the roles of Tectoridin in the proliferation and invasion of human colon cancer cell lines SW480. And to explore its mechanism.Methods1. The MTT assay was used for detection the activity of proliferation inhibition of Tectoridin on SW480 Cancer Cell lines.2. The colony-forming assay was applied for determining the forming of colonies of SW480 tumor cell lines.3. The cell scratch assay was used for detection the migration of tectoridin within 24 h migration of SW480 colon cancer cells.4. We used Western blot assay to research tectoridin on SW480 cell proliferation associated protein Clycin D1, C-myc expression and apoptosis-related protein Bax expression.Results1. By MTT assay, we found that the Tectoridin could significantly inhibit the proliferation of SW480 cells in a time-and- dose-dependent manner(P <0.05).2. The forming of colonies of SW480 cancer cell lines were apparently inhibite by the derivative Tectoridin. We also could find that the higher the concentration of tectoridin, the more pronounced inhibition.3. In cell scratch assay,we found that the Tectoridin could inhibit the migration of SW480 cells in a concentration dependent manner.4. Using Western blot assay showed that Tectoridin can down regulate the proliferation of SW480 cell associated protein clycin D1 and c-myc expression level, and upregulation of SW480 cell apoptosis related protein Bax expression level.Conclusions1. Tectoridin has anti-tumor activity. It can affect the proliferation and migration of SW480 colon cancer cells.2. To up-regulate the expression level of apoptosis-related proteins and promote cell cycle related proteins of Tectoridin on tumor cell maybe the other common mechanisms.
