BackgroundMalignant tumor is a major disease severely threatening human bings for years, and patients with lung cancer occupied a larger proportion of the population. With the development of industry,air pollution and the excessive cigarette consumption, the morbidity and mortality of lung cancer are increasing in each year. According to the distinctions on cell feature and pathology category, lung cancer are mainly classified into non-small cell lung cancer (NSCLC) and small cell lung cancer(SCLC),as NSCLC has essential distinctions with SCLC on origination, survive and transmission, the diagnosis, treatment and prognosis between NSCLC and SCLC are very different in clinic. The scholars abroad have found that members of PAX family widely expressed in the growth of many types of tumor, and it may be associated with the tumors' development and metastases. It has been found that PAX5 highly expressed in B cell lymphoma, in addition PAX5 was found in the expression of neuroblastoma and breast tumor.PAX2 was found expressed in some types of tumor from kidney, bladder,breast, prostate and fallopian tube [2].While the studies about PAX2 and PAX5 in lung cancer are seldom.The immunohistochemistry was used in this research to analysis the expression and the clinical significance of PAX2 and PAX5 in different pathology categories of lung cancer.AimTo investigate the expression of PAX2 and PAX5 in different pathological types of lung cancer and to study the possible effect of PAX2 and PAX5 in the occurrence of lung cancer.Methods1. Collect the bronchial biopsy specimens from patients, between 2014.12.16 and 2015.04.03 of First Affiliated Hospital Of Zhengzhou University, wit h 29 NSCLC specimens,18 SCLC specimens and 13 specimens of pos itive control group. Cases in NSCLC group includes 6 women and 23 m en, age between 35 and 78 years old, average age is 60.28 years old,12 adenocarcinoma cases andl7 squamous carcinoma cases,19 somking cases and 10 non-smoking cases,26 cases with lymph metastase and 3 cases w ith no lymph metastase,16 cases in I+II stage and 13 cases in III+IV sta ge. Cases in SCLC group include 5 women and 13 men, age between 4 8 and 75 years old, average age is 62.56 years old,10 somking cases and 8 non-smoking cases,16 cases with lymph metastase and 2 cases with n o lymph metastase,9 cases in limiting type and 9 cases in diffusing type. Cases in positive control group include 4 women and 9 men, age betw een 25 and 75 years old, average age is 56 years old,12 phlogosis cases and lpulmonary hamartoma. all patients are firstly diagnosed and have ne ver accepted radiation,chemotherapy and other anti-cancer treatment.2. Using the immunohistochemistry to show the expression of PAX2 and PAX5 in 3 different pathology categories of lung cancer, to analysis the distinctions among the expression of PAX2 and PAX5 in 3 different groups and to explore the connection between PAX2,PAX5 and the factors in pathology.3. The judgement of the result:the presence of deep yellow or brown granules in nucleus and cytoplasm means PAX2 positive reaction, PAX5 positive reaction is presented by nucleus staining.5 scopes are randomly selected under the multiplication microscope,signing the staining situation of 200 cells, score according to the density and the proportion of stained cells as no staining (0),pale yellow (1),deep yellow (2),brown (3); the proportion of stained cells<10%(0),10?25%(1),25?50% (2),>50%(3),the final score is the product of density score and proportion score, if the product is not below 3,it means positive staining.4. Statistic analysis:we adopt the statistic software of SPSS 17.0,u sing corrected chi-square test, correlation analysis and Fisher's exact probabilities to deal with qualitative data and using single facto r analysis of variance test to deal with measurement data, Set P< 0.05 for the difference on statistic significance.Results1. The expression of PAX2 in NSCLC is apparently higher than its expression in SCLC and control group.2. The expression of PAX5 in SCLC is apparently higher than its expression in NSCLC and control group.3. In NSCLC group PAX2's positive rate of adenocarcinoma of lung(91.7%) is hi g-her that it of squamous carcinoma(76.5%),but there is on statistic significance o n difference(p=0.343). And there is no difference between lymph node metastasi s group and non- lymph node metastasis group(p=0.068).4. There is no a/parent difference among the tumor stage groups on the expression of PAX5(p=1.00),PAX5's expression in lymph node metastasis group is higher it in non-lymph node metastasis group, and there is statistic significance on this difference(p<0.05).Conclusions1. PAX2 is in high expression in NSCLC group, it maybe associated with the origination of NSCLC, and it can be an indicator for the differential diagnosis between NSCLC and SCLC.2. PAX5 is in high expression in SCLC group, it maybe associated with the origination of SCLC, and it can be an indicator for the differential diagnosis between SCLC and NSCLC.3. PAX5's expression in lymph node metastasis group is higher it in non- lymph node metastasis group, it maybe associated with the transmission of SCLC,and it can be an factor to judge the prognosis of SCLC patients.
