Research objective: To observe the influence mechanism on browning reaction of mice white adipose by an 8-week exercise and resveratrol intervention on 80 Kunming mice as the research objects, so as to explore the effective measures to control body weight under different diet condition.Research methods: 80 mice were randomly divided into 8 groups(n=10): ordinary fodder control group(T1), ordinary fodder and movement group(T2), ordinary fodder + resveratrol group(T3), ordinary fodder and movement + resveratrol group(T4), high-fat fodder control group(H1), high-fat fodder and movement group(H2), high fodder + resveratrol group(H3), and high fodder and movement + resveratrol group(H4). The control group was raised according to different diet conditions, and given lavage of physiological saline according to the regimen; the resveratrol intervention group was given lavage of resveratrol(45 mg/kg/d) according to the regimen; the movement group did swimming exercise according to the scheme and was given lavage of physiological saline according to the regimen; resveratrol + swimming exercise group was given double interventions according to the corresponding requirement. Mice body weight and diet amount were measured during the intervention. After 8 weeks of intervention, blood was taken from mouse heart. was used to detect the concentrations of TG, TC, LDL-C and HDL-C. Changes of hematologic indicators were observed. White adipose tissues were isolated from subcutaneous area, groin, epididymis, and renal surroundings, while brown adipose tissues were isolated from interscapular region. Pathological observation was conducted after paraffin sectioning, HE staining, UCP1 immunohistochemical and frozen section UCP1 immunohistochemical technique, pathological observation; and real-time PCR technology was adopted to analyze the expressions of UCP1, PRDM16, PGC-1?, PPAR? and SIRT1 in adipose tissues.Research results: A 8-week follow-up on changes of mouse body weight and diet amount under high-fat diet condition found that: 6 weeks from the experiment, the increase of body weight in the high-fat fodder control group was significantly higher than that of the normal fodder control group(P<0.05), but there were no statistically significant differences in changes of diet amount(P>0.05); Lee,s index in the high-fat fodder control group(1.71±0.71) was significantly higher than that of the normal fodder control group(1.47±0.51)(P<0.05); A high-fat diet intervention can significantly increase the blood lipid level of mice(P<0.05); exercise and exercise+ resveratrol interventions can significantly reduce the TG level under high-fat diet condition(P<0.01); exercise, resveratrol and exercise+ resveratrol interventions can significantly reduce the TC and LDL-C level under normal diet and high-fat diet conditions, but there were no significant differences among groups(P>0.05); HE staining showed: exercise, resveratrol and exercise+ resveratrol interventions can significantly reduce lipid drops in mice, and increase mitochondrial capillary network; Paraffin section and frozen section UCP1 immunohistochemical displayed: increased UCP1 in subcutaneous white adipose tissue and interscapular brown adipose tissues; RT-PCR results showed that the high-fat control group had lower experiment results than the normal control group, and mRNA expression levels of all experimental groups were higher than the control group at different degrees, but there were no significant differences among groups in SIRT1, UCP1, PPAR?, PGC-1? and PRDM16(P>0.05).Conclusion: Resveratrol intervention and exercise + resveratorl dual intervention both can reduce mouse TG under high-fat diet condition; Resveratrol can increase the UCP1 expressions in subcutaneous white adipose tissue and interscapular brown adipose tissues; Exercise and(or) resveratrol intervention did not have significant effect on mRNA expressions of SIRT1, UCP1, PPAR?, PGC-1? and PRDM16 under different diet conditions.
