Objective To investigate the effect of androgen receptor in cancer-associated fibroblasts (CAF) on the growth and invasive capacities of prostate cancer.Methods We got CAF-ARsi or CAF-sc with knocking down the AR in CAF which has already been immortalized by siRNA. The MTT and soft agAR assay were performed to study the PC3 adherent growth ability and suspended growth ability. To study expression levels of AR and growth factors in CAFs with using the real-time quantitative PCR.Results We cultured and immortalized the resultant cells successfully. The MTT and soft agar assay showed that the PC3 cells cocultured with CAF-ARsi grew slower than the control cells. In the Q-PCR, the expression levels of IGF1, FGF7, FGFIO, SDF1, HGF, and TGFb2 decreased significantly in the CAF-ARsi cells.Conclusion The AR in CAF ceils could promote the growth and invasive capacities of prostate cancer.
