Objectives1 To study the effect of Polygons Multiflori before and after processing medicated serum and main composition on proliferation in BMSCs.2 To study the effect of Polygoni Multiflori before and after processing medicated serum and main composition on the cell cycle in BMSCs3 To study the effect of Polygoni Multiflori before and after processing medicated serum and main composition on the apoptosis in BMSCs4 Study on the Gene level of the mechanism in BMSCs effect of Polygoni Multiflori before and after processing medicated serum and main composition.5 Study on the protein level of the mechanism in BMSCs effect of Polygoni Multiflori before and after processing medicated serum and main composition.Methods1 The proliferation of BMSCs exposed to Polygoni Multiflori before and after processing medicated serum and main composition was analyzed using MTT assay for 24h,48h and 72h.2 The flow cytometry was used to detect the cell cycle changes on BMSCs,which effect of Polygoni Multiflori before and after processing medicated serum and main composition for 24h and 48h.3 The flow cytometry was used to detect the apoptosis on BMSCs,which effect of Polygoni Multiflori before and after processing medicated serum and main composition for 24h and 48h.4 Real-time quantitative PCR(RT-PCR)was used to determine the mRNA expression of hematopoietic genes after the effect of Polygoni Multiflori before and after processing medicated serum and main composition.5 Elisa kit was used to determine the TPO,Kitlg and IL-6 cytokine production of BMSCs effect of Polygoni Multiflori before and after processing medicated serum and main composition.Result1 The Polygonum multiflorum medicated serum has the effect of Promoting BMSCs proliferation,Special the Radix polygoni multiflori medicated serum.Main composition Emodin,Physcion,Rhein and Gallic acid can promote the proliferation on BMSCs,the 5-Hydroxymethyl Furfural dehyde(5-HMF)and Stilbene glucoside inhibite the proliferation on BMSCs.2 Polygoni Multiflori can promote the BMSCs go to the cell cycle active phase,and the Radix polygoni multiflori medicated serum are better.Emodin and Rhein can promote the BMSCs go to the cell cycle active phase,and the Stilbene glucoside inhibite the cell cycle of BMSCs.Physcion,gallic acid and 5-HMF are no significant effect on the cell cycle.3 Radix polygoni multiflori medicated serum can inhibit the apoptosis of BMSCs,and the raw Radix Polygoni Multiflori medicated serum are promoting the apoptosis of BMSCs.The cell apoptosis were significantly decreased after were treated by Emodin and Physcion,and were promoting by Stilbene glucoside.Rhein,gallic acid and 5-HMF are no significant effect on apoptosis4 Polygonum multiflorum medicated serum serum increasing the gene expression of TPO?Kitlg and IL-6,and the Radix polygoni multiflori Preparata medicated serum is higher.Emodin increasing the gene expression of TPO,the Physcion increasing the gene expression of IL-11,and the Rhein increasing the gene expression of IL-11 and Kitlg.mRNA expression of Kitlg were enhanced significantly points after treatment of Stilbene Glucoside,but the Kitlg and IL-6 were weakened significantly points.5 May be the TPO are no expression in BMSCs,so the TPO cytokine can not be detected.The levels of BMSCs factors facilitating Kitlg have the tendency of increased,and administration of 0.1 ?g/ml Physcion was significantly higher than control group.The levels of BMSCs factors facilitating inflammation IL-6 increased more than control group with the Polygoni Multiflori Preparata medicated serum,and the Emodin and Stilbene glucoside less than control group.ConclusionThe blood enriching function of Polygoni Multiflori after processing,beacuse it is closely related with Radix polygoni multiflori medicated serum can promote the proliferation on BMSCs,which the possible action path way of relevant of cell cycle and apoptosis,and the expression of gene and cell secretion related to hemalopoiesis,which might be the mechanism of proliferation of BMSCs.Emodin,Physcion,Rhein and Gallic acid content increases after Polygoni Multiflori processing,these and Radix polygoni multiflorin of BMSCS was consistent with that of proliferation;And the Stilbene glucoside is not displayed on BMSCs proliferation.The result further elucidate the efficacy related changes and processing mechanisms of the Polygoni Multif lori.
