1.Background and significanceFollowed by the increasing population of advanced age,the financial and social burden brought by the cognitive deficits in normal aging process receives wide concerns.The deterioration of age-related cognitive impairment may trigger the incidence of neurodegenerative diseases like Alzheimer's disease.By rescue the cognitive decline among the normal aging of brain,small decreases in conversion rate of age-related cognitive decline to dementia might significantly reduce the prevalence of certain neurodegenerative diseases.In one previous study our lab,it was established that Edaravone may alleviates Alzheimer's disease-type pathologies and cognitive deficits.We intend to design in vivo experiment to test the therapeutic effect of Edaravone on cognitive deficits in aging and explore the related mechanisms2.Materials and methodsC57/6J wild type mouse was picked as a normal aging model.The experiment group was administrated with Edaravone through I.P from 3 to 9 months old while the control group was administrated with NS of same dosage and frequency.Behavioral tests were performed to evaluate the cognitive impairment.Immunofluorescence tests and Western Blot tests were performed to evaluate the synaptic function of the aged brain.To explore the related mechanisms,ELSIA tests and Western Blot tests were performed to evaluate the neuroinflammation and oxidative stress in the aged brain.3.ResultsEdaravone improves cognitive deficits in the aged wild type mouses.It can rescue neuronal and synaptic lose by regulate the expression of synaptic protein.Besides,Edaravone can reduced apoptosis and suppress neuroinflammation and release oxidative stress in aged brain.4.DiscussionThe therapeutic effect of Edaravone on cognitive deficits in normal aging was achieved through its function of reduce the neuroinflammation and oxidative stress.
