Primary Hepatic Carcinoma(PHC),as one of the most common symptoms brought about by malignant tumor worldwide,ranks second place in morbidity and mortality related to malignant tumor in China.As clinical study showed that,a significant increase in serum alpha-fetoprotein(AFP)could be found among about 77~90% of PHC-stricken patients,so AFP was regarded as one of important indicators in early-phase PHC detection and diagnosis.Various methods in clinical testing are currently established in detecting serum AFP up to now: such as chemiluminescence immunoassay(CLIA),time-resolved fluorescence immunoassay(TRFIA),radioimmunoassay(RIA),enzyme-linked immunoassay(ELISA)and GICA detection.However,a great majority of testing equipments and systems involved in the application of these methods are imported,which undoubtedly brings a few of disadvantages in China context: such as complexity of their experimental operations,expensive facilities,high cost of detection etc.All these disadvantages inherent may lead to difficulty in carrying out a large-scale screening of serum AFP indicators for a large number of suspected cancer patients.Therefore,the development of an AFP rapid quantitative detection system seems to be quite important socially and clinically.In the current study,an AFP colloidal gold strip,based on CMOS imaging identification principle,was developed with its designing patterns as follows: a green LED with a wavelength of 540 nm as a light source;a highly integrated CMOS chip as a photoelectric sensor;image recognition of AFP colloidal gold strip by C8051F340 microcontroller chip.The results showed that with detection of four different detection frequencies(5 KHz,30 KHz,50 KHz and 70 KHz),using the image sensor TSL1301 for the determination of the blank strip,the coefficient of variation is below 0.36%;Then mixture with AFP standard sample(25~800 ng/mL),the content of spiked sample of clenbuterol showed a good secondary fitting character with color degree(R2=0.994).AFP recoveries(92.2%~105.5%)and relative standard deviation(RSD)(1.49%~2.25%)showed the method tends to be promising in precision and accuracy.Besides,detecting cost reduced significantly to almost 20% of ELISA kit,which seems more suitable to carry out rapid quantitative screening tests for a large number of blood samples from suspected cancer patients.Adopting CMOS optical imaging identification sensor technology and incorporating RFID RF module in the quantitative analysis of AFP colloidal gold strip applied in the present study present a few of merits: it simplifies the hardware device and makes instruments less sizable and more portable.Meanwhile,this method also makes it more likely to realize intelligent identification of AFP colloidal gold strip and digital management of testing results,which assuredly sheds lights on the solution to quantitative analysis of AFP colloidal gold strip in the process of screening clinical samples. |
