Molecular imaging(MI) is a subject that is used to study the physiological or pathological processes in living organisms in the cell, molecular or genetic level. The technology got great attention and rapid development in recent years. Molecular imaging technology is a method of detecting body. It can get optical image distribution in body noninvasively, continuously and quickly, and expresses some pathological and physiological process with the image in order to provide information for early diagnosis of the diseases. In view of the fact that Biochemical reactions of Bioluminescent Tomography(BLT) in time cannot be controlled and Fluorescence Molecular Tomography(FMT) will bring the interference of Auto fluorescence and stimulated luminescence. Persistent Luminescence material is used for tomography in order to avoid the shortage of those two imaging methods and to achieve the purpose of fault reconstruction. The paper concerns around the fact that the persistent material was used for the reconstruction of biological tomography and explains the controllability and low noise of the Persistent material that used for Biological imaging and the accuracy of Persistent Luminescence Tomography(PLT) reconstruction based on Regularized Orthogonal Least Squares(ROLS) algorithm. The paper mainly include the following:(1)Based on the nature of the Persistent material and inspired by the fact that light can be transmitted through biological tissue and intensity of Persistent material attenuates, we analysis the spectra of stimulated emission and emission of the Persistent material Sr Al2O4:Eu,Dy, have the curve fitting of the attenuation law of the Persistent material, studies the multiple excitation by experiment and finally get the relatively independent time of the multiple excitation for the following work.(2)The paper puts forward the ROLS algorithm was put forward which connects Regularized version of Orthogonal Matching Pursuit(ROMP) algorithm and Orthogonal Least Squares(OLS) algorithm. In the experiments, we got optical information in a relatively independent time on multipoint excitation, and reconstructed the PLT light source based on ROLS algorithm. The experiment and the reconstruction results show that the stronger the light is, the more the signal-to-noise ratio is and the closer the reconstruction results are to the real value. The reconstruction results of the data obtained from the multi-point excitation are better than the results of the single excitation.
