Long noncoding RNAs(lncRNAs)are a new class of RNA which are 200nt to 200kb in length and do not code proteins.LncRNAs regulate gene expression at epigenetical,transcriptional and post-transcriptional levels.LncRNAs have been demonstrated to involve in a broad range of biological and pathological processes through interacting with proteins and/or nuclear acids.Emerging evidence has found aberrant expression of IncRNAs in cancer cells,and demonstrated the crucial roles IncRNAs play in regulation of initiation and development of many cancers including breast cancer.The incidence of breast cancer increased a lot in cities in the last decades.It is becoming the most common cancer among women.The regulation of breast cancer is a complex process involved by many factors and many genes including microRNAs and IncRNAs.For example,the discovery of lncRNA HOTAIR regulation of breast cancer metastasis is regarded as a very important milestone in the fields of either lncRNA or cancer.Different lncRNAs have different mechanisms regulating breast cancer.Clarifying the molecular mechanisms of breast cancer metastasis,and helping establish a reasonable prevention and treatment programs that can greatly improve long-term survival of patients and improve their live quality.In this study,we design the lncRNA primers library by self and conduct quantitative real-time PCR to validate them one by one,developing a high specificity widely detect range and simple breast cancer lncRNA microarray.Using our own design lncRNA microarray to analyze abnormal lncRNA expression in breast cancer cells,then we focus on lncRNA CCAT2 with real time PCR method to verify the results of the chip in 40 breast cancer patients,In addition,we analyzed subcellular localization of lncRNA CCAT2 by nuclear and cytoplasmic extraction.We transfected in vitro to breast cancer cell line MDA-MB-231 overexpression lncRNA CCAT2 and negative control.Finally,we have study deeply IncRNA CCAT2 of the biological behavior in basal-like breast cancer cells MDA-MB-231 such as cell proliferation and migration.(1)Differential expression of IncRNAs in breast cancer cell linesIt is known that lncRNA CCAT2 is downexpressed in breast cancer cell lines the statement of lncRNA microarray and verify the results of the chip in 40breast cancer patients.(2)AnalysislncRNA CCAT2 of subcellular localization(3)LncRNA CCAT2 regulate cell proliferation and migrationMTT assay and Ki67 immunofluorescence method show that the overexpression of lncRNA CCAT2 can inhibit the growth of breast cancer cells compared to negative control,Furthermore,the Wound healing experiments also show lncRNA CCAT2 transfected into MDA-MB-231 cells can significantly inhibit their ability to migrate.In summary,the lncRNA microarray of breast cancer and quantitative real-time PCR confirmed the lncRNA expression was significantly low in breast cancer.In vitro studies confirmed that lncRNA CCAT2 over expression significantly declined proliferation and migration of breast cancer cell MDA-MB-231.Thus,suggesting that lncRNA CCAT2 is potential inhibit tumor in in human breast cancer...
