Expression And Significance Of P16, Ki-67 Combined With Serum SCC-Ag In Cervical Precancerous Lesions And Squamous Carcinoma Of The Cervix

Objective To investigate the expression and significance of p16,Ki-67 combined with serum SCC-Ag in the cervical precancerous lesions and squamous carcinoma of the cervix.Methods The tissue expression of p16,Ki-67 was detected by immunohistochemistry and serum SCC-Ag level before treatment was measured by chemiluinescent microparticle immunoassay in 8 cases of cervicitis,12 cases of low-grade squamous intraepithelial lesions(LSIL),20 cases of high-grade squamous intraepithelial lesions(HSIL)and 20 cases of squamous carcinoma of the cervix(SCC).Results1.The positive rates of p16 in the 4 groups were 0,8.33%,60.00%,95.00%,respectively;There were significant differences between LSIL and HSIL(P <0.05),LSIL and SCC(P <0.05),but no difference between in cervicitis and LSIL(P >0.05),HSIL and SCC(P>0.05).There were no associations with pathological parameters in SCC;2.The positive rates of Ki-67 in the above 4 groups were 0,16.66%,50.00%,100%,respectively.There were significant differences between LSIL and HSIL(P < 0.05),LSIL and SCC(P <0.05),but no difference between cervicitis and LSIL(P >0.05),HSIL and SCC(P >0.05).The expression of Ki-67 in SCC was different in histological grade,the lower the grade,the stronger the positive expression;3.The positive rates of serum SCC-Ag in the above 4 groups were 0,0,25%,75%,respectively.There were no difference in cervical benign lesions(P > 0.05),but there were significant difference between the first three groups and SCC group(P < 0.05).The serum SCC-Ag level was increased with increasing of FIGO stages(P < 0.05).4.Spearman correlation analysis showed that the expressions of p16 and Ki-67 were positively correlated with serum SCC-Ag level in cervical precancerous lesions and SCC,the correlation index were r1 =0.85(P < 0.05),r2 =0.96(P < 0.05),r3=0.86(P < 0.05).Conclusion p16 and Ki-67 is helpful to distinguish benign cervical lesions level,and they both are sensitive to precancerous lesions,but they can not accurately identify cervical precancerous lesions and cervical cancer.If combined with serum SCC-Ag level,they can play a role in the identification of benign and malignant lesions and provide an objective basis for the early diagnosis of SCC.Ki-67 and the serum levels of SCC-Ag were close with malignant degree and clinical stage in SCC.P16,Ki-67 and SCC-Ag may be involved in the pathogenesis of SCC.
