Metformin Inhibit Immune Escape Of Cancer Cells Mediated By Treg Cells Through TGF-? Signaling

Background and Objective:Recent research has shown that AMPK can inhibit tumor metastasis and its mechanism may be related to TGF beta 1,and TGF beta 1 is one of the important cytokine Treg cells differentiations.Previous research has found that TGF beta 1,in the role in the development of tumor were different: this topic through the validation of AMPK inhibiting tumor cell TGF-beta 1 secretion,prevent cancer by Treg cells differentiation is reduced,and eventually results in inhibition of tumor immune escape.Materials and method:1.collect 60 cases in January 2015 to October 2015 in the first affiliated hospital of Nanchang university radical surgery of gastric cancer patients with tumor tissue,made pathological section,using the method of immunohistochemical DAB,observe each group of FOXP3 and the expression of p-AMPK.Both in different stage of relationship of expression in gastric cancer patients By using SPSS 20.0 analysis.2.Different degree of differentiation of gastric cancer cell lines(7901,28,(AGS)cells to metformin treatment,collection and cytoplasm protein on cell cultures,respectively with ELISA and western blot method to detect TGF beta 1 secretion and expression inside the cell.3.The degree of differentiation of gastric cancer cell lines and Jurkat cells trained five days,observation of FOXP3 expression in Jurkat cells.At the same time,build 7901-LKB1 cell lines,and with the Jurkat cell co-culture 0,24,36,48,60 h,with the flow and the IF to observe the expression of FOXP3.Results:1.The degree of lymphatic metastasis of gastric cancer tissues of FOXP3 expression quantity different,from the N0,N1,N2 and N3,the expression of FOXP3 increased gradually.And the expression and p-AMPK present correlation(R = 0.53,p < 0.05).2.As the cell lines increased the degree of differentiation,the secretion of TGF beta 1 was gradually decreased.After treated with metformin,TGF beta 1 secretion decline,intracellular detected TGF beta 1,too.3.Trained the experimental results show that the higher the degree of differentiation of the cells and Jurkat cell co-culture,the lower expression of FOXP3 Jurkat cells;At the same time,7901 and 7901-LKB1 cell lines with Jurkat cells develop,found that the former Jurkat cells FOXP3 expression quantity is higher than the latter.Conclusion:1.The differentiation of Treg cells can promote the development of tumor,AMPK and presents the correlation between expression of FOXP3.2.Metformin can inhibit the expression of TGF beta 1.3.AMPK higher expression can inhibit the expression of TGF beta 1 and secretion,affecting the differentiation of the Treg cells.
