OBJECTIVETo evaluate the effects of MYH-9 as a target for gene therapy in gastric carcinoma.RNA interference(RNAi)was employed to inhibit the expression of MYH-9 in SGC-7901 cells to study the effect on migration and invasion ?METHODSSGC-7901 cells were transfected with MYH-9 si RNA.MYH-9 protein and m RNA levels were detected using Western Blotting and RT-PCR.The migration and invasion assay were used to evaluate the effect of si RNA-mediated inhibition of MYH-9 on invasion and metastasis in gastric carcinoma SGC-7901 cells.RESULTSThe specific si RNA can efficiently block the expression of MYH-9 both at m RNA and protein levels.Compared with the blank control group and negative control group,the expression inhibition rates were 88.9%(t=31.73,P<0.001)and 89.9%(t=33.21,P<0.001)at m RNA level,respectively;The expression inhibition rates were 52.9%(t=11.01,P<0.001)and 60.1%(t=15.53,P<0.001)at protein level,respectively.The result of in vitro migration assay showed that Lenti-MYH9-si RNA cells had significantly reduced invasiveness as compared with negative control cells(t=47.29,P<0.001)and blank control cells(t=46.40,P<0.001).The result of in vitro invasion assay showed that Lenti-MYH9 cells had significantly reduced invasiveness as compared with negative control cells(t=15.53,P<0.001)and blank control cells(t=28.12,P<0.001).CONCLUSIONSLenti-MYH9 can successfully inhibit the expression of m RNA and protein,knockdown significantly reduced the ability of migration and invasion in SGC-7901 cells,MYH-9 might be a good target for gastric cancer treatment.
