Aims To explore the effects of FMT on dextran sodiumsulfate (DSS)-induced colitis model in mice and whether if donor intestinal microbiota concerned. Methods BALB/c male mice were treated with DSS for 7 days, then randomized into four groups:control group, FMT group with autologous feces, FMT group with one donor's feces, FMT group with multi-donors'feces. Control group and FMT groups were given normal saline (NS) or homogenate (made by fecal and NS) gavage respectively, homogenate were stored in the fridge at-80 ? for 1-2 weeks. Body weight and DAI scores of mice in each group were recorded. Changes in distal colon were studied by histopathology. Bacterial species in feces were also studied. Results FMT caused reversal of body weight, DAI scores and histopathological scores of colon in FMT groups mice and the reversal was more remarkable than in control group. Balance of gut microbiota were perturbed by DSS and then were altered by FMT. Gut microbiota of FMT group with autologous fecal were the most similar to it before DSS treated. The mucosal layer of FMT groups showed nearly normal gross structure,but there were still more inflammatory cells of FMT group with one donor's fecal and FMT group with multi-donors'fecal than FMT group with autologous fecal. There no significant differences were observed in effect between FMT group with one donor's fecal and FMT group with multi-donors'fecal.Conclusions FMT using frozen stool is effective in dextran sodiumsulfate (DSS)-induced colitis model in mice through its modulation on gut microbiota. Effects of FMT using autologous feces is better than using healthy donors'feces, suggesting that effects of FMT DSS-induced colitis model in mice were concerned with donors'intestinal microbial communities, the one more similar to mice when they were health has better effects. FMT using one donor's feces or multi-donors' feces has the same effects.
