Objective This study aimed to investigate mRNA expression of Epidermal Growth Factor Receptor (EGFR) and Vascular Endothelial Growth Factor Receptor-2 (VEGFR-2) gene mutation in non-small cell lung cancer (NSCLC) patients and summarize the relationship between the expressions and clinicopathologic features of non-small cell lung cancer patients; to analyze the gene expression correlation and explore the mechanism of EGFR signaling pathway and VEGF/VEGFR signaling pathway, in order to provide theoretical evidence of combined application of anti-epidermal growth factor receptor and anti-angiogenesis targeted drugs for NSCLC patients.Method A total of 128 NSCLC patients admitted to the Department of Thoracic Surgery of General Hospital of PLA between 2014 and 2015 were included, and their clinical data including age, gender, smoking history, pathological type, differentiation and TNM staging were recorded. EGFR E18-21 mutations and mRNA expression of EGFR and VEGFR-2 gene in fresh specimens of tumor tissue or CT-guided percutaneous needle biopsy of lung were quantified by using quid chip technology, then the acquired data were statistically analyzed with SPSS19.0 software to observe if there is any correlation.Result Among the included patients, the rate of EGFR gene mutations was 47.7%, including 21.1% E19 gene mutation,26.6% E21 gene mutation,0% E18 and E20 gene mutations. There was a significant correlation between EGFR E19 gene mutation and smoking history (r=0.238, p<0.01); It showed EGFR E21 gene mutation had a correlation with tumor histological type (r=-0.213, p<0.05) and the degree of tumor differentiation (r=-0.190, p<0.05). EGFR gene expression associated with tumor type (r=0.244, p<0.01). VEGFR-2 gene expression showed a correlation with pathological type of (r=0.271, p<0.01) and tumor differentiation (r=0.237, p<0.01). There was a significant correlation between EGFR and VEGFR-2 expressions (r=0.320, p<0.01).Conclusion EGFR mutations tend to occur in exons 19 and 21. wherein E19 mutation likely occurs in patents with no smoking history and E21 mutation occurs in high-differentiated adenocarcinoma patients. EGFR and VEGFR-2 gene expressions are higher in adenocarcinoma patients, and VEGFR-2 gene expression levels positively correlated with tumor differentiation degree. There is co-expression of EGFR gene and VEGFR-2 gene.
