BackgroundViral hepatitis is a worldwide epidemic of infectious diseases, which is a serious hazard to human health. There are millions of people infected with viral hepatitis every year. Nearly 1 million 500 thousand people die of this disease every year. According to the latest data released by the World Health Organization 2012: the world has 20 million people infected with hepatitis B virus, of which there are 2.4 million people with chronic hepatitis B virus infection and the HBV surface antigen positive about 1.2 billion people, If the pathogenic factor is not removed, with the development of the disease, chronic hepatitis B will appear to varying degrees of fibrosis, long-term development will inevitably lead to cirrhosis of the liver. At present, the main methods of diagnosis of liver fibrosis are percutaneous liver biopsy, but because of its inability to dynamically observe the dynamic changes of liver fibrosis and the effect of treatment, it is limited in clinical practice.Recent studies have shown that Tissue dispersion quantitative analysis technique of ultrasound elasticity imaging effect in the inspection and monitoring of liver fibrosis significantly, but its clinical value for liver fibrosis is still a lack of effective experimental demonstration.In this study, using real-time tissue elastography ultrasound quantitative tissue diffusion analysis is to detect rat models with different stages of liver fibrosis, and observed by comparing tissue diffusion quantitative techniques, and contrast with the liver biopsy results, and explore the application value of tissue dispersion quantitative analysis in rat liver fibrosis stages and provide a reference for clinical non-invasive diagnosis of liver fibrosis. PurposesTo explore its application value in the classification of liver fibrosis, and to provide a preliminary reference for clinical noninvasive diagnosis of liver fibrosis with dispersion quantitative analysis technique. Objectives and methods1 objectives72 SPF 8 weeks old male Wistar rats(Shandong Lukang Pharmaceutical Co., Ltd., license number: LSCXK-20130001) and body mass of 240~268g average(250.2±9.1) g. 64 rats were selected as the experimental group by random digits table, and the other 8 were as control group.2 Methods72 male Wistar rats were randomly divided into experimental group( n= 64)and control group( n= 8), The experimental groups of rats were gavaged with the volume fraction of 60% CCl4 olive oil solution to form different stages of liver fibrosis, the control group with normal saline.all the rat underwent tissue dispersion quantitative analysis to obtain 12 elastic parameters,these above parameters in rats of different liver fibrosis stages were compared,then the correlation with pathological stages were analyzed. ResultsExcept for COMP,ASM,CORR,there were significant differences of all parameters in rats of different liver fibrosis( P <0.05),and were correlated with pathological stages( P <0.05).among these parameters,%AREA had the highest related coefficient( r = 0.891, P =0.001), The ROC curve was made by %AREA to estimate the fibrosis stage.the area under ROC curve for %AREA was 0.914(? the control group+ S0), 0.963(?early liver fibrosis S1 + S2), 0.969(?middle liver fibrosis S3), 0.948(early cirrhosis S4),respectively. ConclusionsThe technique of tissue dispersion quantitative analysis has a good evaluation value of liver fibrosis stages,It is able to provide a preliminary reference for clinical non-invasive diagnosis of liver fibrosis.
