Objective:To investigate the effects of diosgenin on monocrotaline-induced pulmonary hypertension in rats and the expression of nuclear tra-nscription factor kappa B (NF-?B) in lung tissue.Methods:36 Sprague-Dawley (SD) rats were fed adaptively for 1 week and then were randomly divided into the following three groups normal control group (control group), MCT-induced model group (model group), MCT-induced with Dio intervention group (intervention group), each group has twelve rats. The control group given a single intraperitoneal injection the same conditions of saline; the others were injected with monocrotaline (60mg/kg) to induce PAH. From day 1 to day 28, control group and model group were respectively given the same dose of vehicle(5%ethanol+distilled water), and intervention group were given Dio 80mg/kg (dissolved in 0.5ml vehicle) fed to intervene. At 29 days, we measured the mean pulmonary artery pressure under general anesthesia, then dissected the rat, separate the right ventricular(RV) and left ventricular+septum(LV+S), and calculated the right ventricular hypertrophy index [RV/(LV+S)]; We observed the morphological changes of small pulmonary arteries,and measured the counts of inflammatory cell ,arteriole wall thickness/vascular external diameter (WT%), and vascular area/total vascular area (WA%). We observed the expression of lung tissue NF-?B in rat by immunohistochemical staining; and then compared the results among groups.Results:1.The determination of hemodynamic:compared with the control group, mPAP and RV/(LV+S) were significantly higher in model group(P<0.01). After the intervention of diosgenin, mPAP and RV/(LV+S) were significantly lower in intervention group than in model group (P<0.01).2.Pulmonary artery morphology:compared with the control group, the counts of inflammatory cell, WT% and WA% were significantly increased in model rats (P<0.01). After the intervention of diosgenin, the counts of inflammatory cell, WT% and WA% were significantly lower in intervention group than in model group (P<0.01). 3.The expression rate of NF-?B in lung tissue of rats:compared with the control group, the expression rate of NF-?B was significantly increased in model rats (P<0.01). After the intervention of diosgenin, the expression rate of NF-?B was significantly lower in intervention group than in model group (P<0.01).4.Correlation analysis:the expression rate of NF-?B were positive correlated with the counts of inflammatory cell and WT% and WA% of pulmonary small arteries (P<0.01).Conclusion:1.Diosgenin could reduce monocrotaline-induced pul-monary hypertension in rats; 2. Diosgenin could reduce the expression of NF-?B in rats induced by monocrotaline, mitigate inflammatory response and improve pulmonary vascular remodeling.
