The Reaseach On Baicalin For The Treatment Of Hypertension Disorder Complicating Pregnancy

Part1 literature studies about the Traditional Chinese medicine on the treatment of pregnancy-induced hypertension syndromeObjective and Methods:Pregnancy-induced hypertension syndrome belongs to pre-eclamsia,gestational vertigo and gestational anasarca area in traditional Chinese medicine. This article summarize the reportes in recent 26 years about the traditional Chinese medicine to treat pregnancy-induced hypertension syndrome.Make a simple statistical analysis of TCM Syndrome Types about pregnancy-induced hypertensi-on syndrome, analyze the frequency of traditional Chinese medicine which has been widely used in the disease of pregnancy-induced hypertension syndrome.Results and Conclusions: Through this study, we find out nine of themost common syndrome type in TCM from No.1to No.9and twenty of the most widely used herbs in the treatment of hypertension disorder complicating pregnancy.the syndrome type in TCM are,in order,syndrome of water diffusion due to spleen deficiency, syndrome of exuberance of liver fire, syndrome of upper hyperactivity of liver yang,gestational vertigo with syndrome of spleen deficiency and liver hyperactivity,syndrome of stagnant dampness-heat, spleen yang-deficiency and kidney yang-deficiency syndrome,syndrome of orifices confused by phlegm, syndrome of yin deficiency of liver and kidney,syndrome of deficiency of both qi and blood.The herbs are,inorder,tuckahoe,atractylodes,angelia,astragalpreparations,amomum villosum, alisma, plantain Seed,chinese yam,orange peel,keel,oyster,herba taxilli,lycium barbarum,radix paeoniae alba,gastrodia elata,uncaria,scutellaria baicalensis,chrysanthemum,fructus corni,eucommia ulmoides oliv.Through this study, we can make TCM Syndrome Types unified and use the herbs which seems more effective in the treatment of pregnancy-induced hypertension syndrome, make the TCM Syndrome Types standardized and make the herbs which will be used unified.Part2 Effects of baicalin on the expression of mitogen-activated protein kinases in Human Umbilieal Vaseular Endothelial Cells induced by serum of PreeclampsiaObjective:According to the results of the first part of the research of traditional Chinese medicine,we find that baicalin was widely used in the treatment of Preeclampsia,now we also find that the component of radix scutellariae can protect the endothelial cells.In recent years, The study found that MAPK signaling pathway can mediate inflammatory reaction and it is an important signal transduction pathways in inflammatory factor expression. It may be involved in the injury of endothelial cell in the pathogenesis of preeclampsia. Baicalin is one of the main effective components of Scutellaria Baicalensis which is belong to origanum. It can anti-microbico,anti-infection,anti-inflammatory,antioxidant.pro tect the blood vessels of the heart and brain.The study found that anti-inflammatory effect of baicalin may be related to inhibit the phosphorylation of MAPK signal pathway level and raise the expression of anti-inflammatory cytokine. We cultivate human umbilical vein endothelial cells, build model of the damage of endothelial cell. Study the effect of baicalin on the MAPK signaling pathways and the disease of Preeclampsia, to provide the oretical basis for the future widely applications of baicalin.Methods: We cultivate endotheliocytes of human umbilical vein and divide them into different groups. The first group :The control group which has three branches,they are normal pregnancy group(NP group),mild preeclampsia group(MPE group)and severe preeclampsia group(SPE group).Interventions: cultivate HUVECS with modified eagle Medium(D-MEM)(1X), liquid(high glucose) and serum of normal pregnancy women,mild preeclampsia women and severe prepreeclampsia women,after 48 hours we collect this cells.RT-PCR and Western blotting were used to measure the m RNA and protein expression levels of ICAM-1,phosphorylated P38 and phosphory-l ated JNK in HUVECs.The second group also known as the inhibitor group, There are two inhibitors in this group. It has four branches.Interventions: P38 inhibitor groups: pretreated HUVECS with high glucose D-MEM and respectively serum of mild preeclampsia women or severe preeclampsia women,after 48 hours,then deal this two group cells with SB203580 for 24 hours. Then collect this cells.JNK inhibitor groups: According to the method as mention-ed above,after 48 hours,then deal this two group cells with AEG3482 for 24 hours.The expression of P38,JNK and m RNA was measured by real-ti me PCR, Western blotting was employed to measure the expression of phosphorylated P38 protein,phosphorylation of JNK protein and ICAM-1 Protein in the treated cells.The third group consists of three groups. In the environment of high glucose,pretreated cells with Serum of normal pregnancy women,mild preeclampsia women and severe preeclampsia women for48 hours,,then deal this three group with baicalin for 24 hours.then the cells were harvested for the further exploration.The expression of ICAM-1,P38 and JNK m RN A was measured by real-ti me PCR, Western blotting was employed to measure the expression of phosphorylated P38 protein, phosphorylation of JNK protein and ICAM-1 Protein in the treated cells.Results: 1.compared to NP group, the expression of protein of phosphorylated P38, phosphorylation of JNK and ICAM-1 and the expression of m RNA of ICAM-1, P38 and JNK has increased significantly in HUVECS in the group of MPE and SPE. The expression of protein m RNA of the P38, JNK and ICAM-1are higher than MPE group.These results have statistical significance.2. the comparation of inhibitor group and the frist group: compared with MPE group and SPE group,the phosphorylation of p38 and ICAM-1 protein expression level and the m RNA expression level of the P38 inhibitor groupsis Significantly lower.compared with MPE group and SPE group,the phosphorylation of JNK and ICAM-1 protein expression level and the m RNA expression level of the JNK inhibitor groups is Significantly lower.3.Blood-serum + baicalin groups: the protein expression level of phosphorylation of P38 and the phosphorylation of JNK and ICAM-1 and the m RNA expression level of the JNK in MPE+ baicalin group and MPE+ baicalin group are lower than MPE and SPE groups.But the protein expression level of phosphorylation of P38 and the phosphorylation of JNK and ICAM-1 and the m RNA expression level of NP+baicalin group compared with NP group,there was no significant difference.Conclusions: The Blood-serum of PE can stimulate the activation of p38 MAPK,JNK/SAPK pathway and increase the expression of ICAM-1in human umbilical vein endothelial cells, the strength of the influence associated with the severity of the illness, with the increase of chronic,the protein expression level of phosphorylation of P38 and the phosphorylation of JNK and ICAM-1and the m RNA expression level of the JNK,p38 and ICAM-1 increased clearly.This experiments show us that baicalin can significantly inhibit the expression of ICAM –1 which has adverse effects on endothelial cells, we owe this infiuence to baicalin can inhibit JNK/SAPK and P38 MAPK signaling pathways. So we draw the conclusion that baicalin has a protective effect on endothelial cells.
