Objective In this research, with super stimulate ovulation control mice as the research object, using the technology of reverse transcriotion-polymerase chain reaction(RT-PCR) and western blot, to observe the effect of eletroacupuncture on IGF-1 expression in the endometrium of controlled ovarian hyperstimulatio mice,and observing the best interventional time of eletroacupuncture through statistics and contrast.Methods The female mice were randomly divided into natural cycle group,COH group, down-regulation group,Gn group,HCG group and embryo transfer group. COH group and EA group are establish the model of long-term superovulation mice: Daily intraperitoneally injection of triptorelin 0.02ug/g at 9am,for 9 days,and injection of Pregnant Mare Serum Gonadotropin(PMSG) 10 IU at 6pm on day 9; after 46-48 hours,injecting human chorionic gonadotropin(HCG) 10 IU. Down-regulation group,Gn group,HCG group and embryo transfer group were interviewed the therapy of eletroacupuncture at the day of beginning down-regulation,injecting PMSG,injecting HCG and transplantation respectively. Acupuncturing Guan Yuan,Zhong Ji and San Yinjiao at the same time of the day for 15 min, 1 times / day.Mating with male mice after injection of HCG,then collecting cell, culturing and transplanting.Result1. After controlled ovarian hyperstimulation,the pregnancy rate of COH group is significantly lower than natural cycle group,with statistical significance(p<0.01);After being therapied by eletroacupuncture,the pregnancy rate of Gn group,HCG group and embryo transfer group are all higher than COH group, with statistical significance(p<0.01);HCG and embryo transfer group had the best effect,compared with Gn group,with statistical significance(p<0.01);After being therapied by eletroacupuncture,the pregnancy rate of down-regulation group reduce instead,compared with COH group,with statistical significance(p<0.01).2. After controlled ovarian hyperstimulation,the average implantation sites of COH group is significantly lower than natural cycle group, with statistical significance(p<0.01);after the treatment of eletroacupuncture,the implantation sites of HCG group is the highest in electroacupuncture group,and is significantly higher than COH group, with statistical significance(p<0.01);but the group of down-regulation,Gn and embryo transfer's have not been improved.3. The endometrium of each group has been detected IGF-1m RNA, after controlled ovarian hyperstimulation,the expression of COH group has significantly reduced,compared with natural cycle group, with statistical significance(p<0.05);after the the treatment of eletroacupuncture,the group of HCG and embryo transfer have significantly increased, compared with COH group, with statistical significance(p<0.05).4. The endometrium of each group has been detected IGF-1 protein. after controlled ovarian hyperstimulation, the expression of COH group has significantly reduced,compared with natural cycle group, with statistical significance(p<0.05); after the the treatment of eletroacupuncture,the each group with electroaupunture has the higher expression than COH group, with statistical significance(p<0.05); HCG group has the highest expression, compared with natural cycle group, with statistical significance(p<0.05).Conclusion1. Eletroacupuncture can influence the implantation rate of embryo transplantation, the implantation rate will be the highest when the the therapy were interviewed after the injection of HCG.2. IGF-1 participated in the process of embryo implantation.3. Eletroacupuncture can influence the IGF-1 expression on endometrium in the he process of embryo implantation,and the relative expression of IGF-1 wil be the highest when the the therapy were interviewed after the injection of HCG.
