In the 21 st century,with the improvement of people's living standards,the extension of the average life expectancy,the spread of unhealthy lifestyles,the pace of people's life becomes faster and the working pressure increases,then the major threat to human health has changed,mentalillness has become one of the maindis eases of the threat to modern human health.Anxiety disorder has become a common psychological disorder in clinical practice,it is one of the hot spots in the current treatment of mental disease to find and develop the anti-anxiety drugs from traditional Chinese medicine(TCM).In the 2015 edition of "Chinese Pharmacopoeia",Cortex Albizziaehave effects of relieving depression and soothing the nerves,commonly used in uneasy,depression,insomnia and other symptoms in clinic.Therefore,it is important to study on Cortex Albizziae.On the basis of our previous studies,a preliminary study on the serum pharmaeochemistry and the chemical composition of the anti-anxiety extracts from Cortex Alibizziae has been done,which preliminary explore the material basis of anti-anxiety.For further study on the serum pharmacochemistry of the an ti-anxiety extracts from Cortex Alibizziae,the study of Characteristic spectrumof the anti-anxiety extracts in vitro from Cortex Alibizziae should be done firstly.By adopting the technology of UPLC,Acqu ityTMUPLC BEHC18 chromatographic column(50 mm x 2.1 mm,1.7?m),flow rate of 0.3 ml/min.the 10 batches of the anti-anxiety extracts from Cortex Alibizziae are analyzed,to establish a method of characteristic spectrum of it,the chrom atographic conditions were optimized,the gradient of mobile phaseand the precision of the instrument,repeatability were was investigated,and calibrate13 common peaks of characteristic spectrum.The peak of character istic spectrum of the anti-anxiety extracts from Cortex Alibizziae were assigned by using the method of UPL C-Q-TOF-MS.As a result,the RSD of the relative retention time and relative peak area of common peak were in limit range,the resolution of each chromatographic peak was better,the reproducibility and good stability show that this method is accurate and reliable,which lays the foundation for the study of serum pharmaceutical chemistry of the late.To study the serum pharmacochemistry of the anti-anxiety extracts from Cortex Alibizziae by using the method of UPLC-Q-TOF-MS.AcquityTMUPLC BEHC18 chromatographic column(50 mm x 2.1mm,1.7?m),the dissolution of the test sample,the elution system,the column temperature,the precipitation method of serum protein were examined,blood was collected in four time points after ad ministration of 15 min,30min,1H and 2h.By comparing the charac teristic spectrum of drug-containing serum,blank serum andthe anti-anxiety extracts from Cortex Alibizziae,With MasslynxV 4.1 MS workstation,five transitional components in blood were identi fied preliminarily;most of them are lignan glycosides :glaberi de I;(-)-syringaresinol-4-4'-bis-O-?-D-glucopyranoside;(-)-syringaresinol-4-o-?-D-glucopyranoside;5,5'-dimethoxy-7-oxol ariciresinol;(-)-syringaresinolTo seperate chemical compositions of the anti-anxiety extracts from Cortex Alibizziae by using silicagel column chromatography,low pressure preparative chromatography and thin-layer preparative chromatography technology,MS?1H-NMR?13C-NMR were used to identify the structure of these chemical compositions,six compounds from the anti-anxiety extracts from Cortex Alibizziae are as follows:(-)-syringaresinol-4-o-?-D-glucopyranoside;2,6-dim ethoxy-4-hydroxyphenol-1-O-D-glucopyranoside;Daidzin;(-)-syring aresinol-4-o-?-D-apiofuransyl-(1?2)-?-D-glucopyranoside;(-)-syringaresinol;(-)-syringar esinol-4-4'-bis-O-?-D-glucopyrano side.
