Objective:With the change of the environment and many factors such as dietary habit,the incidence of colon cancer in China increase rapidly at the rate of 3.33%, colon cancer has been a serious threat to the health of the residents in our country. Colon cancer incidence and mortality are rising along with the age growth. Due to the tendency of population ageing in our country, the research on the pathogenesis of colon cancer and early diagnosis become a new challenge. After Vitamin D combining with Vitamin D Receptor?VDR?, it work on the specific DNA sequences to control the expression of the gene.Systematic studies have confirmed that VDR is expressed abnormally in many tumors such as colon cancer, pancreatic cancer, esophageal cancer, ovarian cancer, liver cancer, lung cancer and so on.The anticarcinogenic mechanism of vitamin D on colon cancer is still not very clear, it remains to be elucidated. The correlation between vitamin D and cancer in the recent research mainly around the mediating specific signaling pathways,controlling cell cycle, and regulating the body's immune.Wnt signaling transduction pathways have been associated with dozens of high-risk cancers like breast cancer, cervical cancer,stomach cancer, thyroid cancer, liver cancer and so on. It has been reported that the abnormal of Wnt signaling pathway exist in 90% of early-stage colorectal cancer. Asthe most common pathology type of colon cancer, the pathogenesis of colonic carcinoma has received extensive attention of scholars at home and abroad.Most of the previous researches on the relationship of VDR and ? chain protein??-catenin?expression are animal experiment.Researches on VDR and ?-catenin expression in human colon adenocarcinoma are rare at home and abroad. In this article, the expression of VDR and ?-catenin in 65 cases of colonic adenocarcinoma tissue and corresponding adjacent normal tissues?more than 5 cm far from the tumor edge?are tested by immunohistochemistry method to evaluate their roles in the colon adenocarcinoma pathogenesis.Methods:1. Using immunohistochemical method to check the VDR and ?-catenin expression in colonic adenocarcinoma tissue and corresponding adjacent normal tissues?more than 5 cm far from the tumor edge?, and analyze the relationship among the expression level and clinical factors like gender, age, lymph node metastasis,differentiation degree.2. Using SPSS19.0 statistical software to analyze immunohistochemical results between groups by chi-square test. When it does not meet the conditions of chi-square?theoretical frequency < 1?,selecting the Fisher's exact test. Indicators were analyzed by the Spearman rank correlation analysis, adopting bilateral inspection, inspection level ?= 0.05.When P <0.05, difference was considered statistically significant.Results:1. After 65 cases of colon adenocarcinoma tissue and corresponding adjacent normal tissue immunohistochemical results were analyzed, the strong positive expression rate of VDR in colon adenocarcinoma was 9.23%?6/65? when the strong positive expression rate in the normal tissue adjacent to carcinoma was 35.38%?23/65? with significant differences??2= 12.83, P < 0.01?.2. The abnormal expression of VDR has nothing to do with the sex or age of colon adenocarcinoma patients?P > 0.05?, it is related to the degree of differentiation,lymph node metastasis and serosa infiltration?P < 0.05?.3. After 65 cases of colon adenocarcinoma tissue and corresponding adjacent normal tissue immunohistochemical results were analyzed, the abnormal expression of ?-catenin in colon adenocarcinoma was 98.46%?64/65?when the abnormal expression rate in the normal tissue adjacent to carcinoma was 4.62%?3/65? with significant differences??2= 114.60, P <0.01?.4. The abnormal expression of ?-catenin has nothing to do with the gender, age,differentiation of colon adenocarcinoma?P > 0.05?, it is related to lymph node metastasis and serosa infiltration?P < 0.05?.5. In the correlation analysis between the abnormal expression of ?-catenin and abnormal expression of VDR in the colon adenocarcinoma tissue: r = 0.67, P < 0.01,it can be seen in colon adenocarcinoma that the abnormal expression of ?-catenin and abnormal expression of VDR were positive correlation.Conclusions:1. The abnormal expression of VDR has nothing to do with the sex or age of colon adenocarcinoma patients, it is related to the degree of differentiation, lymph node metastasis and serosa infiltration, the abnormal expression of VDR may be associated with the development and metastasis of colon adenocarcinoma.2. The abnormal expression of ?-catenin has nothing to do with the age, gender,differentiation of colon adenocarcinoma, it is related to lymph node metastasis and serosa infiltration, the abnormal expression of ?-catenin may be associated with the development and metastasis of colon adenocarcinoma.3. The abnormal expression of ?-catenin and abnormal expression of VDR were positive correlation in colon adenocarcinoma. The results suggested that the lower expression of VDR makes vitamin D and VDR composites decrease, may change the?-catenin, activate Wnt pathways, lead to the occurrence of colon adenocarcinomawith many other factors. Combing detection of ?-catenin and VDR may has important implications for the diagnosis and prognosis of colon adenocarcinoma.
