This study used pressure volume loop analysis method to explore the Cuscuta chinensis Lam. Flavonoids(CCLF) of rat cardiac hemodynamic effects. Consists of two parts of the experiment: the purpose of experiment one: To explore the Cuscuta chinensis Lam. Flavonoids impacted on cardiac hemodynamics in vivo rats of the preload acute reduced model. Methods: 30 Sprague-Dawley rats were randomly divided into three groups(n=10): sham operation group(SOG), operation group(OG), Cuscuta chinensis Lam. Flavonoids(CCLF) 200 mg/kg group.(1)SOG: Amount of saline once a day in the first 7 days, on the eighth day, apply Powerlab 16 physiological recorder and Millar catheter to detect hemodynamic index, open the abdominal cavity, isolate the inferior vena cava(IVC), cross the surgical suture and ligation, indicate no ligation point in time. Then, detect hemodynamic index for 30 min;(2)OG(the preload acute lower model group): Amount of saline once a day in the first 7 days, on the eighth day, apply Powerlab 16 physiological recorder and Millar catheter to detect hemodynamic index, open the abdominal cavity, isolate the IVC, cross the surgical suture and ligation, indicate ligation point in time. and detect hemodynamic index 30 min;(3)Amount of CCLF once a day in the first 7 days. complete the same steps as OG. Results:(1)CCLF could improve the ESP, dP/dt max, dP/dt mix, SW, CO, SV, EF, SW/EDV, dP/dt max/EDV, EDV, dP/dt min indexes of the preload acute reduce model rats and reduce the HR, EDP, Tau indexes of the rats.(2)CCLF could improve the ESPVR and reduce EDPVR indexes of the preload acute reduce model rats.(3)CCLF could improve the PVA and CE indexes of the preload acute reduce model rats. Conclusion: CCLF could improve the cardiac systolic and diastolic funcation, promote the energy utilization efficiency, enhance the heart blood pump out and return ability, and ensure the body's normal blood supply of the preload acute reduce model rats. The purpose of experiment two: The establishment of the in vitro rat working heart and the pressure-volume loop guidance. Methods: 20 SD rats were randomly divided into three groups(n=10): control group(CK), isoprenaline group(ISO).(1)CK: In the working heart model without any intervention, the hemodynamic parameters were recorded for 50 min by using the Powerlab 16 channel physiological recorder and the Millar catheter.(2)ISO: When work heart were recorded 10 min, 1 ?mol/L ISO was added for anterograde perfusion and continue to record 40 min. Conclusion:(1)Millar catheter through the aorta retrograde perfusion cannula method was used to establish in vitro work heart model success;(2)Compared with control group, the pressure-volume loop of ISO group was moved to the left and expanded, verify the ISO positive inotropic effect, also showed that the model could satisfy the pressure-volume loop method in vitro cardiac hemodynamics experimental requirements.
