Objective:To explore the effects of suppressing CHD1L(Chromodomain helicase/ATPase DNA binding protein 1- like) gene expression by CHD1 Lsi RNA on h MLH1(human mut L homolog 1) genes expression in hepatoma carcinoma cell line SMMC7721 and proliferation or migration and apoptosis of hepatoma carcinoma cell line SMMC7721.Methods:1.After down-regulated the expression of CHD1 L in hepatocellular carcinoma cell line SMMC7721 by transfecting CHD1 Lsi RNA, detect the expression of m RNA and protein of h MLH1.2.After transfection, the CCK8 experiment, apoptosis and Transwell migration experiment are adopted to observe the proliferation, apoptosis and migration of hepatocellular carcinoma cell line SMMC7721.3.Statistical analysis was performed by using SPSS17.0 statistical software,measurement data was represented by using(x±S), all the experimental data were compared by two independent samples t-test analysis, P<0.05, the difference was statistically significant.Results:1.Down-regulated the expression of CHD1 L in hepatocellular carcinoma cell line SMMC7721, the m RNA and protein expression levels of h MLH1 rised.2.Down-regulated the expression of CHD1 L in hepatocellular carcinoma cell line SMMC7721, the proliferation and migration of hepatocellular carcinoma cell line SMMC7721 are inhibited and its apoptosis is promoted.Conclusion:1.In the hepatocellular carcinoma cell line SMMC7721, down-regulating the expression of CHD1 L may promte the expression of h MLH1.2.In the hepatocellular carcinoma cell line SMMC7721, down-regulating the expression of CHD1 L inhibit the proliferation and migration of hepatocellular carcinoma cell and promte its apoptosis...
