BackgroundPost-stroke depression(PSD)is one of the most common complications of the patients with cerebral apoplexy.PSD is a kind of mood disorder caused by stroke,and it can be associated with Cognitive impairment.Immune and inflammatory response is an important post-stroke pathology,which directly affects the central nervous system damage and repair.Immune and inflammatory response is directly related to long-term prognosis and complications of depression,cognitive impairment,etc.Treg is negative immune regulatory role cells.Treg play a protective role in the brain after cerebral ischemia inflammation,and is closely related to the depression.The changes of Treg expression may affect immune imbalance and the occurrence of PSD.ObjectivesLearn Regulatory T cells state in peripheral blood of PSD rats,and explore its effect on the immune status and progression of the PSD.Methods1.Prepare focal cerebral ischemia rat model by middle cerebral artery obstruction(MCAO)technique.Each focal cerebral ischemia rat model was raised in a single cage,and was given chronic unpredictable mild stimulation(CUMS)to prepare the PSD rat model.2.Evaluate the nerve function defect degree of the focal cerebral ischemia rats model by Longa method.Observed experimental rats weight changes by measuring the weight.Test rats the changes of interest and happy feeling by sucrose water consumption.Test rats the changes of spontaneous movement ability andexploration ability by open field test(OFT),through the data to assess depressive state of the model.3.Test CD3+ CD4+ CD25+ Foxp3+ regulatory T cells level in peripheral blood of rats in each group by flow cytometry and analyze its significance.Results1.The results of behavior tests(1)Compare with the sham operation control group,the weight of the PSD and depression group showed slow growth,at the 21 st day of the stress,compare PSD and depression group rats weight with the sham operation control group,the differences had statistical significance(F = 27.525,P = 0.000),compare the depression group rats weight with PSD group,there was no statistical significance(P > 0.05);(2)After 7 days,14 days,21 days of the stress,the sucrose water consumption of PSD and depression group rats was lower than the sham operation control group,the differences had statistical significance(P < 0.01),after 14 days,21 days of the stress,compare PSD group with the depression group,there was no sstatistical significance(P >0.05),after 21 days of the stress,compare the simple stroke group with the sham operation control group,there was no statistical significance(P > 0.05);(3)After 7 days,14 days,21 days of the Stress,the OFT horizontal motion distances of PSD and depression group rats were lower than the sham operation control group,the differences had statistical significance(P < 0.01),compare the depression group with PSD group,there was no statistical significance(P > 0.05),compare the sham operation control group with the simple stroke group,there was no statistical significance(P > 0.05);(4)After 7 days,14 days,21 days of the stress,OFT vertical movement numbers of PSD and depression group rats were lower than the sham operation control group,the differences had statistical significance(P < 0.01),after 14 days,21 days of the stress,compare the simple stroke group with the sham operation control group,there was no statistical significance(P > 0.05).2 The results of flow cytometric tests7 days after MCAO,the expression of CD3 + CD4 + CD25 + Foxp3 + Treg cells in peripheral blood of simple stroke group,depression group and PSD group rats decreased,compare with the sham operation control group,the differences had statistical significance(P < 0.01);the expression of CD3 + CD4 + CD25 + Foxp3 + Treg cells in the peripheral blood of Simple stroke group was higher than 7 days after MCAO group,the differences had statistical significance(P < 0.01);compare the expression of CD3 + CD4 +CD25 + Foxp3 + Treg cells in the peripheral blood of PSD group with 7 days after MCAO group,there was no statistical significance(P > 0.05),compare PSD group with the simple stroke group,there was no statistical significance(P > 0.05).ConclusionsCD3+ CD4+ CD25+ Foxp3+ regulatory T cells level in peripheral blood of PSD rat model decreased,imbalance of regulatory T cells is very likely to play an important role in the immunological injury of Post-stroke depression,regulatory T cells may be involved in the pathogenesis of Post-stroke depression.This conclusion provides a reference to further clarify the immune mechanism of post-stroke depression.
