| ObjectiveOral cancer cell mtDNA damage,repair,dynamic changes in the copy number induced by exogenous oxidation stimulation;the relationship between mitochondrial DNA damage and cell activity,mitochondrial DNA damage and nuclear DNA damage;to exploration the oral cancer pathogenic mechanism and prevention methods.MethodsSelect the high incidence and malignant degree of strong oral squamous cell carcinoma SCC-25 cell lines as experimental model,choose the skin fibroblast BJ strain as control model experiment,with four concentrations of H2O2(120 μ M-960 μM)and six time(15min,1H,2h,24 h,48h,72h)induced cell response to oxidative damage,mtDNA damage,repair,dynamic changes in the copy number was determined by the method of ss-q PCR;MTT method for determination of cell activity and growth inhibition;nuclear DNA fracture was observed by H2 AX.ResultsBy mtDNA damage in the two kinds of cells ss-qPCR method for the detection of,dynamic changes of repair found that cells SCC-25.Under the low level of oxidative stress,oxidative damage and repair response,copy number will not change;high levels of stimulation,the formation of irreversible damage,as time continues,copy number significantly decreased.For normal BJ cells and oxidation of low levels of stimulation,and did not cause cell damage repair mechanisms,with increasing levels of stimulation they exhibit damage and a certain degree of repair.It can be seen,cancer cells to oxidative stress is moresensitive Respond more strongly.At the same time,two kinds of cells was detected by MTT assay,cell carcinoma LC50 concentration was only 0.5 times higher than that of the control cells,the cancer cells under low concentration stimulated when oxidative damage corresponding to.When the fault detection of cancer cells nuclear DNA and found that oxidation stimulated under,will also lead to nuclear DNA fragmentation.ConclusionsThis experiment by using ss-qPCR experimental techniques on oral cancer cells to detect the mtDNA,the cell oxidative damage,repair and copy number changes in the dynamic change were obvious;combined with cell growth inhibition detection and cell’s nuclear DNA fragmentation situation analysis.Results show that,oral cancer cells showed the more sensitive response to oxidative stress and oxidative stress as a major cancer causing factors,will provide a new way for the early prevention and treatment of oral cancer. |
