Objective1.To observe the treatment of thymosin to asthma model rats.2.To explore the regulation role of thymosin for Th1/Th2 in asthma model rats respectively.3.To explore the expression level of T-bet ?GATA-3?IL-4?IFN-? in asthma model rats respectively.Methods1.60 SPF grade male rats(6-8 w)were randomly divided into six gro ups:control group? asthma model group,dexamethasone(0.5 mg/kg)gro up,thymosin high dose group(5 mg/kg),thymosin middle dose group(2.5 mg/kg),thymosin low dose group(1.25 mg/kg).The asthma model was con structed with ovalbumin.The rat in the control group and asthma model grou p were injected with normal saline by intraperitoneal,while other rats were injec ted with corresponding drugs.Then measure the tide volume,respiratory fre quency,calculate ventilation in each rat.2.The method of ELISA was used to detect the expression of IL-4 a nd IFN-? in the serum of rats.3.lung tissues pathology morphology was observed by using HE stai ning.4.The method of immunohistochemical was used to detect the protei n expression level of GATA-3 and T-bet factor in the lung tissue in each group.5.The method of western blot was used to detect protein expression level of GATA-3 and T-bet factor in the lung tissue in each group.Results1.It measured the tide volume,respiratory frequency,calculate ventilation in each rat(P<0.05).2.The concentration of IL-4 in the serum was higher and IFN-? was lower in the asthma model group than the other groups(P<0.05).3.Normal group did not see eosinophils and inflammation in lung tissue.Asthma model group can see a lot of lung tissue eosinophil infiltration in the red dye,pathological changes,such as epithelial cell injury.After treatment with thymosin and dexamethasone,the above phenomenon was significantly reduced.4.The protein expression level of GATA-3 was higher and T-bet was lower in the asthma model group than the other groups by the method of mmunohistochemistry,and there are significant difference(P<0.05).5.The protein expression level of GATA-3 was higher and T-bet was lower in the asthma model group than the other groups by the method of Western blot,and there are significant difference(P<0.05).Conclusion1.Thymosin had therapeutic effect on asthma rats.2.Thymosin upregulation of IFN-? may be achieved by upregulating T-bet.3.Thymosin inhibition of IL-4 may be achieved by downregulating GA TA-3.
