Regulation Of Icariin On Immune Cell Function In Mice

ObjectiveIn order to research the immune enhancement pharmacological effects of icariin,in this paper,a preliminary immune activity screening model was established,in which mice splenocytes,peritoneal macrophages and dendritic cells derived from bone marrow were chosen as the target cells.We briefly discuss the immune regulating function of icariin at the cellular level.MethodsPrimitive cultured mice splenocytes,peritoneal macrophages and dendritic cells derived from bone marrow were treated as the target cells.MTT method was used to detect the influence of icariin on proliferation of lymphocyte and lymphocyte infected by Staphylococcus aureus,and the effect of macrophage phagocytosis.We used ELISA method to detect the influence of icariin on cytokines secretion of lymphocytes,lymphocyte infected by Staphylococcus aureus and dendritic cells.Flow cytometry was used to detect the influence of icariin on cell cycle of splenocytes.ResultsStimulating lymphocyte by icariin could enhance lymphocyte stimulation indices.Stimulating lymphocyte by icariin with concanavalin A(Con A)or lipopolysaccharide(LPS)could also enhance lymphocyte stimulation indices.The latter performanced better.The stimulation of lymphocytes with icariin could increase the secretion of IL-2,IL-4,IL-10 and IL-2 in lymphocytes,decrease the percentage of G1 phase in the cell cycle,and increase the percentage of lymphocytes in the S phase.Icariin was used to stimulate bone marrow dendritic cells,which was able to enhance the secretion of IL-2,IL-10 and IL-12P70 in dendritic cells.Stimulating lymphocyte which was infected by Staphylococcus aureus by icariin could enhance lymphocyte stimulation indices.Compared with the bacteria infection control the secretion was increased,but compared with icariin control the secretion was decreased.Icariin could enhance macrophage phagocytosis of Staphylococcus aureus.ConclusionIn some extent dose,the icariin can promote lymphocyte proliferation and LPS or Con A stimulation of lymphocyte proliferation and Th1 and Th2 cytokines secretion in lymphatic cells.Increase the percentage of cells entering the S phase,which can improve the immune regulation activity of lymphocytes.Icariin being used to stimulate dendritic cells can promote the secretion of IL-2,IL-10 and IL-12P70.After lymphocytes are infected by Staphylococcus aureus,icariin can promote lymphocyte proliferation and Th1 and Th2 cytokines secretion,improve the phagocytosis of macrophage,which verify the icariin can regulate the function of immune cells.
