Objective: our aim is observe the effect of Compound barbata granules on the MAPK/ERK-PI3K/Akt signal transduction pathway in liver fibrosis,and find out the function of Compound barbata granules in the treatment of rat's liver fibrosis.Methods: 84 SPF male SD rats were randomLy dividing into 7 groups,including normal control group(group A),model group(group B),Colchicine positive control group(group C),Rhubarb Zhechong pill group(group D),Compound barbata granules high dose,medium dose,low dose group(group E,F,G),a total of seven groups,12 rats in each group.Using the method of composite factors intervention to prapared the hepatic fibrosis rat model by 40% CCl4,they were treated as follows: control group free diet,normal diet;The rats of model group were injected CCl4 in intraperitoneal regularly according to weight,with high fat and low protein diet and 15% alcohol fed rats,then began to gavage by normal saline after successful modeling;the rats of Colchicine positive control group,Rhubarb Zhechong pill group,Compound barbata granules high dose,medium dose,low dose group were gavaged by corresponding drugs 4W after successful modeling.When the treatments are over,collect all of the specimens.Using the experimental methods of pathological sections of HE staining,immunohistochemical staining,Western blot and RT-PCR detection to detect these specimens and to observe the MAPK/ERK-PI3K/Akt signal transduction pathway in the expression of various indicators.Results: 1.Compared with control group,in the rest of the groups the liver index was increased,the difference was statistically significant(P<0.05);compared with model group,the liver index in treatment each group was decreased,the difference was satistically significant(P<0.05).In Compound barbata granules high dose,medium dose,low dose group,the liver index in high dose group was decreased significantly,the difference was statistically significant(P<0.01);2.Under the light microscope,in normal group,liver tissue morphology was normal;model group and Compound barbata granules low dose group had obvious changes of hepatic histology and a certain degree of liver injury and fibrosis;the liver tissue of Colchicine positive control group,Rhubarb Zhechong pill group,Compound barbata granules high dose and medium dose groups have different degree of liver fibrosis performance;3.Compared with the control group,the p42/44 MAPK,p38MAPK,PI3 K,Akt expression volume ofrat liver tissue of model group and treatment each group were higher,the difference was statistically significant(P<0.05);Compared with the treatment each group,the p42/44 MAPK,p38MAPK,PI3 K,Akt expression volume of rat liver tissue of model group were lower,the difference was statistically significant(P<0.05);Compared with the rest of treatment each groups,the p42/44 MAPK,p38MAPK,PI3 K,Akt expression volume of rat liver tissue of Compound barbata granules high dose group were lower,the difference was statistically significant(P<0.05);Compared with the Compound barbata granules low dose group,the p42/44 MAPK,p38MAPK,PI3 K,Akt expression volume of rat liver tissue of the medium dose group were lower,the difference was statistically significant(P<0.05).4.Compared with the rest of groups,the p42/44 MAPK,p38MAPK,PI3 K protein expression volume of normal group were lower,the difference was statistically significant(P<0.05);Compared with the rest of groups except Compound barbata granules high dose group,the Akt protein expression volume of normal group were lower,the difference was statistically significant(P<0.05);Compared with the each treatment group except Compound barbata granules low dose group,the p42/44 MAPK,Akt protein expression volume of control group were lower,the difference was statistically significant(P<0.05);Compared with the each treatment group,the p38 MAPK,PI3K protein expression volume of controlgroup were higher,the difference was statistically significant(P<0.05);Compared with the rest of each treatment group,the p42/44 MAP K,p38 MAPK,PI3K protein expression volume of Compound barbata granules high dose group were lower,the difference was statistically significant(P<0.05);Compared with Compound barbata granules low dose group,the p42/44 MAPK,p38MAPK,PI3 K protein expression volume of Compound barbata granules medium dose group were low er,the difference was statistically significant(P<0.05);Compared with each other,the Akt protein expression volume of each treatment group do not have significant difference(P>0.05).5.Compared with control group,the PI3 K m RNA,Akt m RNA expression volume of normal group were lower,the difference was statistically significant(P<0.05);Compared with Rhubarb Zhechong pill group,Compound barbata granules high dose,medium dose group,the PI3 Km RNA expression volume of control group were higher,the difference was statistically significant(P<0.05);Compared with Compound barbata granules medium dose,low dose group,the PI3 Km RNA,Akt m RNA expression volume of Compound barbata granules high dose group were lower,the difference was statistically significant(P<0.05);Comp ared with Compound barbata granules low dose group,the PI3 Km RNA expression volume of Compound barbata granules medium dose group were lower,the difference was statistically significant(P<0.05);Compared with the each treatment group,the Akt m RNA expression volume of control group were higher,the difference was statistically significant(P<0.05);Compared with both sides,the Akt mRNA expression volume of Compound barbata granules medium dose group and low dose group do not have significant difference(P>0.05).Conclusion: Compound barbata granules may inhibiting the activatio n of HSC or accelerate exhaustion of HSC through inhibiting the MAPK/ERK-PI3K/Akt signal transduction pathway conduction,so as to reduce the degree of fibrosis in the rat model of CCl4 induced liver fibrosis rats,and improve the hepatic fibrosis in rats.
