Objective:This experiment which mainly by reagent to induce myocardial fibrosis,aimed to observe the expression of TRPC channel of each subtybe before and after the activation of heart fiber cell;to understand the function of TRPC channel in fibrosis reaction.Finally,to find out a new way to prevent and heal the dieases related of myocardial fibrosis.Methods: 10 SD rats were randomly divided into 2 groups,each group of five,namely the intervention group(group A)and blank control group(group B),for A set of experimental group rats by intraperitoneal injection of one-time isopropyl adrenaline(dose: 5 mg/kg.D);Group B blank control group rats by intraperitoneal injection of one-time saline(dose: 5 mg/kg.D),continuous injection of 14 d,take two groups of rats cardiac apex of myocardial,1 ?Take a few of both group of rats myocardial,each rat make two pathological section,to compelate HE staining and Masson staining respectively,then observed fibrosis degree and the collagen fiber distribution of myocardial tissue under electron microscope,compared to the experimental group and the blank control group in the changes of myocardial tissue.2? Take the apex of the heart tissue in rats of group A,purification and cultivation of fiber cells,Obtain the homology of myocardial fiber cells.3?It can be divided into M group and N group,group M as ck,the N group was given the reagent stimulation of TGF?1,after the stimulus,extract the m RNA of collagen fiber of the both two groups.To observe the transcription of the purpose gene,give the PCR detection,get the result of the expression of the purpose gene,then in order to verify the PCR results,give western blot examination.aim to further validation of correlation in protein translation level.Results: 1.Through the way of HE staining and Masson staining,to contrapose the fibrosis degree of the apex of myocardial tissue in both two group.The results all suggest that it exist significant differences.Group A is significantly higher than group B,it shows obviously statistical significance.For fiber cells of group A and group B,PCR result shows below:TRPC1 in group A and B,no significant difference(P > 0.05);TRPC2,false gene;TRPC3 gene exists obvious difference(P < 0.05);TRPC4,no obvious difference(P > 0.05);TRPC5 has no obvious difference(P > 0.05);TRPC6 gene exists obvious difference(P < 0.05)TRPC7 has no obvious difference(P > 0.05);To sum up,in fibrosis models,The m RNA expression of TRPC3 and TRPC6 appears obvious difference.Using Western Blot technique to further verify the result of PCR,TRPC3 and TRPC6 m RNA translation has significant correlation(P<0.05),consistent with the results of PCR.Conclusion: After giving stimulation,myocardial fibroblasts activated into myofibroblast.As a result of the existence of cascading waterfalls amplification reaction in fibrosis,the degree of fibrosis and the area is extensive.In the process of fibrosis,TRPC channel subtypes in transcription and translation level exist high expression,especially the TRPC3 and the TRPC6.They are the most significant in particular.This study indicates that the myocardial fibrosis and The expression of TRPC channels are relevant,with statistical significance.It has statistical significance.It can be used as a new target to intervene myocardial fibrosis,to control the process of fibrosis cascade.Aimed to provide theoretical basis for anti fibrosis drug research and development.
