| Objective:Taking Suanzaoren Decoction at Mao Shi and You Shi timing,observe the effect of improving cognitive function of rats in Alzheimer's disease,then explore the mechanism.Methods:Choosing 48 elderly male SD rats,then were randomly divided into normal group,model group,Mao Shi group,You Shi group.D-galactose and A?1-42 were taken to make model of AD rats.At 6:00 and 18:00,Mao Shi and You Shi groups were respectively gavaged of Suanzaoren Decoction 10.5g per kg and normal groups at 6:00 every day were gavaged of gastric volume of physiological saline for 42 days.At 42 th day,Morris water maze test was begun,temperature and autonomic activity of rat,and the concentration of5-HT?MT?MTR from serum Elisa method were measured,HE staining detection showed rat hippocampus morphology,immunohistochemical method show Taup Ser199 loci positive expression in hippocampal CA1 area.Results:(1)In the navigation test of Morris test,The time of model group to collision was obvious higher than normal group(P<0.05),The time of maoshi group and youshi group to collision were lower than model group(P<0.05),The time of maoshi group to collision was obvious lower than youshi group(P<0.05),In the spatial probe test,times that rats went Through the hidden platform in one minutes,model group were less than normal group(P<0.05),maoshi group and youshi group were more than model group(P<0.05),maoshi group were more than youshi group(P<0.05).(2)The concentration of5-HT?MT?MTR showed model group was lower than normal(P<0.05),maoshi group and youshi group were higher than model group(P<0.05),maoshi group was higher than youshi group(P<0.05).(3)HE staining: cellular structure of normal group is clear,Big round,colored clearly,neuron cells are close and neat.Cellular structure of model group is fuzzy,nucleus are swelling and fragmentation,shape is irregular,a large number of neuron are degeneration,nuclear is pyknosis.cellular structure of Mao Shi group and You Shi are clear,the nucleus are swelling and fragmentation less.Morphological rules,the neurons are arranged more neatly,Mao Shi group is closer to the normal group.(4)Immunohistochemical: neuronal cytoplasm of model group rats in hippocampal CA1 area are tan,positive cells were significant higher than normal group(P<0.05),neuronal cytoplasm of Mao Shi group and You Shi rats in hippocampal CA1 area are light yellow,positive cells were significant lower than the model group(P<0.05),the positive cells,Mao Shi group was obvious lower than You Shi group(P<0.05).Conclusion:(1)Cognitive function of rats in Alzheimer's diseaseof taking Suanzao ren Decoction with suitable time get good effect.(2)Compared Mao Shi group and You Shi group in the cognitive function rats,Mao Shi group is better than that of You Shi(3)With the raise of the concentration of the serotonin(5-HT)? melatonin(MT)? melatonin receptor(MTR),AD rats sleep is improved,so as to improve the symptoms of cognitive function in AD rats.(4)Suanzaoren Decoction can effectively reduce the the hippocampus Taup S199 positive cells expression rate of model group and delay the extent of excessive Tau protein phosphorylation,which in turn improves cognitive function in model group rats. |
