Aim: To research the pharmacological treatment and mechanism of Danggui-ShaoyaoSan(DSS) on pregnancy induced hypertension. To evaluate the function of DSS on rats model with pregnancy induced hypertension, in the blood pressure, the 24 h urine protein, placental tissue pathology morphology and liver and renal function as incaditor.Discussion from arginine vasopressin(AVP) and nitric oxide(NO) pathway Danggui-shaoyao-san therapeutic effects of pregnancy-induced hypertension, with a view to Dangguishaoyaosan the clinical treatment of hypertensive disorders of pregnancy provide an experimental basis.Method:(1) The method of nitric oxide synthase inhibitor(L-NAME) is adopted to establish the rat model of pregnancy induced hypertension.(2)Measuring rat tail artery mean arterial pressure with automatic noninvasive blood pressure measuring instrument.(3)Metabolic cages were used to collect 24 hours urine of rats, 24 h urine protein content was measured by BCA method.(4)The Placental pathological changes were observed by HE staining.(5)The levels of serum ALT, AST, TP, ALB, GLB, BUN, Cr were detected by the automatic biochemistry analyzer. PT was evaluated by the blood coagulation analyzer.(6)The level of K+?Na+?Cl-were detected by the automatic biochemistry analyzer.(7)The plasma level of AVP, Ang?, ET-1, PGI2, TXA2, ?-HCG and E were detected by enzyme-linked immunosorbent assay(ELISA).(8)The relative expression of(AVP)V2R ?e NOS protein in renal tissue and VEGF protein in placental tissue were detected by Western Blotting.Result:(1) Rat gestation 11 day intraperitoneal injection of L-NAME 25mg/kg, to pregnancy 18 day, reproducible and stable gestational hypertension model.(2)Compared with the model group, Danggui-shaoyao-san was significantly lower average pulse pressure, decrease 24 h urinary protein significantly.(3) Placenta syncytiotrophoblast nodules decreased trophoblast compensatory value can be improved in DSS group.(4)The serum ALT, AST, TP, ALB, GLB, BUN, Cr levels were improved to varying degrees, significantly lower plasma PT in DSS groups.(5) The serumlevel of K+?Na+?Cl-increased significantly.(6)The plasma level of AVP, Ang?, ET-1 significantly reduced in DSS groups.(7)The relative expression of(AVP)V2R protein were detected and e NOS protein were increased in renal tissue by Western Blotting.The relative expression of VEGF protein were detected in placental tissue by Western Blotting.Conclusion:The stable model of the pregnancy induced hypertension rats could reproduced by the method of L-NAME. DSS can reduce the blood pressure significantly of pregnancy-induced hypertension in rats, reduce 24 h urinary protein content, improve liver and kidney function, slow the pathological process, while corrective electrolyte imbalance to treat pregnancy induced hypertension.
