Clinic study1.1 Objective: To study the distribution law of traditional Chinese medicine Syndromes of Bronchial Asthma and give a basis for TCM syndrome differentiation and the clinical treatmentthe,the study was carried out through the TCM syndrome survey of 142 patients.1.2 Methods:Feference formulation asthma syndrome table,and combined with the asthma control test(ACT) score and asthma control table, analysis and sum up the data of 142 cases of patients with asthma using statistical software,to summarize the distribution regularity of TCM syndromes of Bronchial Asthma.1.3 Results: There are 65 patients have cold asthma(accounting for 45.77%)among the 142 cases of patients.And patients asthma susceptibility are mainly with Yang deficiency and phlegm accounting for 35.91%.It illustrates that there is some kind of relationship between the incidence of asthma and physical factors.2. Experimental study:1.1 Objective: To explore the immune mechanism and molecular of Xiaochuanping granule in treating asthma,we observed the effect that of xiaochuanping granule on cytokine IL-17 and Notch signaling pathway in asthmatic rat model,and to explore the mechanism and molecular.1.2 Methods:Divide the 84 male SD rats into 7 groups. There are The control group,Asthma model group, Prednisone group,Jinshuibao group,Xiaochuanping powder low-dose group,Xiaochuanping powder dose group,Xiaochuanping powder high dose gro up. 12 rats in each group. After adaptive feed 7days began modeling.Sensitization after 2 times, with ovalbumin(Ovalbumin, OVA) spray excitation, 1 times a day, 5weeks of excitation. End of the modeling,Traditional Chinese medicine group was given Xiaochuanping granules,western medicine group given prednisone and Chinese medicine group be Jinshuibao,normal group and model group were given the same amount of normal saline,once a day for 2 weeks.Every groups with Xiaochuanping granule(high dose group was 0.625g/kg/d,equivalent to amount of adult dosage 12.5times,middle dosage group was 0.313g/kg/d,equivalent to amount of adult dosage6.25 times,low dose group was 0.157g/kg/d,equivalent to amount of adult dosage 3.13times) and Jinshuibao group(0.313g/kg/d) to fill the stomach.After two weeks,The sampling was made to detect the content of IL-17 in serum and Bronch-oalveolar lavage fluid, and the expression of Notch1-4 in Notch signaling pathway, and to explore the mechanism of Xiaochuanping granule in the treatment of asthma.1.3 Results:1.3.1 The content IL-17 of Rat serum:Compared with the normal group,the content of IL-17 in serum of rats in model group were significantly higher than the normal group(P<0.01). Compared with the model group, each treatment group rats serum IL-17 content were decreased(P<0.01).Compared with the traditional Chinese medicine Western medicine group and high dose group, the difference was not statistically significant(P>0.05). Compared with Chinese medicine group and low dose of Chinese medicine group, the difference was not statistically significant(P>0.05). The content of IL-17 in the serum of Chinese medicine high dose group and high dose group was lower than that of traditional Chinese medicine group(P<0.05).1.3.2 The content IL-17 in bronchoalveolar lavage fluid of rats:Compared with the normal group, the content of IL-17 in model group was significantly higher than that in normal group(P<0.01). Compared with model group,the content of IL-17 in bronchoalveolar lavage fluid was decreased in all treatment groups(P<0.01). There was no significant difference between western medicine group and traditional Chinese medicine group(P>0.05). There was no significant difference between the traditional Chinese medicine group and the low dose group(P>0.05). The content of IL-17 in bronchoalveolar lavage fluid was lower than that in traditional Chinese medicine group in Chinese traditional medicine group and high dose group(P<0.05).1.3.3 The measuring results of Notch1、Notch2、Notch3、Notch4 in rat lung tissue.Compared with the normal group, the model group of lung tissue Notch2 and Notch1 were significantly higher than the normal group(P<0.01). Compared with the model group, all the medication groups have lower Notch2 and Notch1 expression in rat lung tissue(P<0.01),There was no significant difference between western medicine group and traditional Chinese medicine group(P>0.05),Compared with the low dose of traditional Chinese medicine in traditional Chinese medicine, the expression of Notch1 and Notch2 are decreased(P<0.05). There was no significant difference between the traditional Chinese medicine group and the low dose Chinese medicine group(P>0.0 5).Compared with the normal group, the average optical density of Notch3 was higher than that of the normal group,the average light density of Notch3 in lung tissue were significantly decreased, and there was no significant difference between the two groups(P>0.0 5). There was was not significantly increased of Notch4 expression in all groups.1.3.4 Detecting the level of Notch1-4 mRNA expressed in lung tissue of rats by RT-PCR:Compared with the normal group,the expression of Notch 1 and Notch 2 of the lung tissue in model group was significantly higher(P<0.01).Compared with the model group,the expression of Notch1 and Notch2 m RNA of the lung tissue in treatment group were decreased( P<0.05). There was no significant difference between the high dose traditional Chinese medicine group and the western medicine group(P>0.05). Compared with the low dose of traditional Chinese medicine group,the expression of Notch1 and Notch2 m RNA in the middle dose of traditional Chinese medicine group are decreased(P<0.05). Compared with the low dose of traditional Chinese medicine group, the expression of Notch1 and Notch2 m RNA in Chinese patent medicine group are decreased(P<0.05).The expression of Notch3 m RNA in asthma model group and control group all decreased, there was no significant differences between them(P>0.05). The expression of Notch4 m RNA in lung tissue of model group and control group were not obvious.2. Conclusion:2.1 Cold asthma is the most common type of TCM syndrome in the majority of 142 cases of asthma patients.2.2 Immunological pathogenesis of asthmatic may be inhibition of leukocyte interleukin-17(IL-17) produced,which could alleviate the airway inflammation.And down-regulation of the expression of Notch signal pathway of Notch1 and Notch2,To inhibit the Notch signaling pathway to participate in the differentiation and activation of T cells, and to delay airway remodeling of asthma. |
